项目摘要

Reproductive success during early pregnancy relies on complex and progressive interactions between the maternal genital tract, gametes, fertilised oocytes and embryos. Our basic research interest is to understand the roles of reactive oxygen species (ROS : nitric oxide (NO), hydrogen peroxide (H2O2) etc..) and their neutralisation by antioxidant systems in female reproductive functions. The physiological roles in the female tract of ROS and antioxidant defenses are poorly understood and it is our global goal to understand these roles. The first objective of the research program is to determine 1) which ROS or antioxidant defenses are involved in the physiological process of fertilisation and embryo implantation? 2) How ROS and antioxidants are regulated in the female tract in relation to the physiological events of fertilisation and implantation? 3) How ROS and antioxidant defenses affect prostaglandins biosynthesis? Prostaglandins are involved in many physiological events in reproduction. The study of the expression of ROS generators and antioxidant enzymes in the bovine female tract according to the physiologic status has first been investigated by us. This project requires the analysis of protein and genetic expression in the uterus and the oviduct of the cow. Investigation of the type of cells involved is also an important objective. In vitro studies with uterine and oviductal cells are also planned. This research will lead to a better understanding of the physiology of reproduction of a wide spread domestic animal in Canada, the cow. This research will be tentatively transpose or eventually compared to other species. This research can also help us to diagnose infertility and finally help us to improve fertility in domestic animals.
