The biomanufacturing of retroviral and lentiviral vectors

The production of retroviral vectors for large-scale clinical trials is limited due to the adherent nature of the existing packaging cell lines. Furthermore, serum used to supplement media can contribute to introduce biological contaminants like proteins, viruses and prions. Serum is also an expensive raw material and it increases the complexity, duration and cost of downstream processing (purification, concentration and infectious virus recovery). We are proposing to generate retroviral but also lentiviral packaging cell lines that could grow in suspension and serum-free media (SFM). Cells with such properties could allow the biomanufacturing of vectors in bioreactors with practically no size limit.
