Increasing gene dosage provides a powerful means of probing cellular functions, such as growth or cell division. The sophisticated molecular genetic methods available with yeast, makes this organism an ideal model system for studying fundamental cellular functions. In total, yeast cells have ~6000 different genes and a subset of ~1000 of these genes are each essential because their deletion kills the cell. We developed a novel system to identify yeast genes whose increased activity can override and suppress the lethality associated with the loss-of-function of a given essential gene. Comprehensive mapping of these genetic interactions will generate a functional wiring diagram of the cell because it defines genes that work together to control essential cellular functions. Because many cellular functions are conserved from yeast to human, the resultant genetic network will be informative about genetics underlying human diseases and it will be useful for the development of new therapeutics.
