Functional synergy between the DNA topology and DNA repair machineries during the initiation of DNA replication

A fundamental component of many normal and disease-related processes is the replication of DNA, the genetic material of all living cells. Despite its importance and the discovery of many molecules that are involved in it over the years, the regulation of DNA replication is still largely unknown. In previous studies, Dr. Zannis-Hadjopoulos' team identified Ku, a multifunctional protein involved in DNA repair, as involved also in the regulation of DNA replication through its interaction with other proteins, among them topoisomerase II, a protein that is involved in maintaining the correct architecture of the DNA. They now propose to analyze the manner by which Ku interacts with topoisomerase II and examine the mechanism by which this protein complex is involved in DNA replication.
