This project will investigate bioprocessing criteria which are important to the development of innovative bio-manufacturing applications. The overall objective is to elucidate and characterize the equipment and operational conditions of a novel pre-sterilized disposable-plastic "bag" design of bioreactors for production of pharmaceuticals and other biological products. At present, relatively little fundamental bioprocessing information is available for these bioreactor types, especially for a second-generation design (introduced within the past two years) which are of special interest in this project because its capability (via two-dimensional rocking-shaking motion intensification of a flat bag) of improved aeration-agitation conditions. The study would quantify the mixing and aeration criteria for a range of microbial fermentation applications, thereby extending current knowledge now limited to animal-cell culture systems. Relevant process engineering correlations between the systems and operational variables will be established based on relatively inexpensive physically-simulated fermentation cultures.
