Staphylococcus aureus is an opportunistic pathogen that causes a variety of diseases in humans and animals. In dairy industry, S. aureus mastitis is extremely difficult to control and usually develops into a chronic infection, at least partially due to invasion of bacteria into epithelial cells, which may help resist defense mechanisms of the host and decrease the efficacy of antibiotic treatment. The global objective of this proposed research is to continue my previous NSERC discovery grant and to study the mechanisms by which surface adhesins of S. aureus attach to and invade bovine mammary epithelial cells, using Clf-A as a model system. Invasion of bovine mammary epithelial cells by S. aureus have been reported in primary and cultured mammary epithelial cells and in a mouse model system. Adherence and the subsequent invasion processes are the two most important and decisive prerequisites to the establishment of chronic S. aureus intramammary infections. These two processes are mediated by a large, structurally related, family of surface-expressed proteins including clumping factors A. This proposed research will focus on Clf-A.
The surface-located Clf-A has an unusual dipeptide repeat linking the ligand binding domain to the
wall-anchored region. Our recent research has shown the potential of the repeats for genotying. We also have identified a putative Clf-A receptor on cultured mammary epithelial cells. Further, we have constructed Lactobacillus casei ATCC 393 carrying Clf-A with different numbers of the repeats. In this new proposal, we will further confirm and characterize the Clf-A receptors on both cultured and primary mammary epithelial cells, determine the effect of varying lengths of the repeats on the binding to epithelial cells and effect on pathogenesis of S. aureus in a mouse model, and explore the signaling pathways after Clf-A binding with the receptor on the epithelial cells. The techniques and the strategies used in this study will be adopted in the future for studying other adhesins. It is hoped that these studies will enhance our understanding of pathogenic mechanisms of S. aureus and consequently provide new strategies for a better prevention and therapy program.
