Infectious diseases have become a major threat to public health, causing more than 13 million deaths per year, of which
approximately 50% occur in developing countries. Clinicians rely on the laboratory to identify the pathogen responsible for
these diseases and initiate proper treatment, which can take days. Thus, to reduce the health burden of these diseases, a
rapid and accurate diagnosis is required. We propose to develop a universal diagnostic platform for point-of-need (PON)
diagnostics (performed where they are needed), which can be used to rapidly identify infectious diseases from nasal swabs,
blood or urine with little hands-on time that can be performed by non-experts. Our device consists of three main
components; first, a specimen delivery system, which isolates the virus or bacteria for downstream applications; second, a
sample preparation system, which releases genetic material from the pathogens for detection; and third, a DNA amplification
and detection component to identify specific pathogen-associated genes. Using these three components, we can identify any
pathogen-of-interest in a PON device in less than 30 minutes, which can be employed by physicians, dentists, veterinarians,
food-processing plants, and also for at-home use. The successful development of this diagnostic platform will have a
significant impact on the management of infectious diseases worldwide.
