L'arthrose demeure la maladie chronique la plus répandue des animaux de compagnie. On estime que 20% des chiens et chats adultes en souffrent en Amérique du Nord et que sa prévalence atteint 80% chez les animaux âgés. Pourtant malgré des décennies de recherches pour diminuer la progression de la maladie ou au moins soulager la douleur des animaux atteints, nous devons constater l'absence de progrès dans cette lutte contre l'arthrose. La seule explication de ce déficit récurrent dans le développement d'agents anti-arthrose réside dans la faible validité des modèles et des variables utilisés pour reproduire la maladie et évaluer ses atteintes.Le Groupe de recherche en pharmacologie animale du Québec (GREPAQ) en partenariat avec ArthroLab Inc. a comme premier objectif de valider un nouveau modèle d'arthrose expérimentale chez le rat. Dans la validation de ce modèle, nous établirons le profil des atteintes de la fonction locomotrice, ainsi que deux mesures de la sensibilisation du système nerveux récemment validées par notre groupe chez le chat arthrosique. De plus, nous établirons des bio-marqueurs génétiques et de neuro-transmetteurs/-modulateurs sur la base de nos travaux antérieurs. Dans un second temps, nous procèderons à la translation des marqueurs validés sur ce modèle vers les atteintes arthrosiques naturelles chez le chien et le chat.La validation de ce modèle novateur d'arthrose cumulera les atteintes structurelles et la douleur chronique sur le petit animal de recherche. Ceci permettra une approche novatrice de la maladie, collée à sa pathogénèse et son expression comportementale, et positionnera ArthroLab Inc. comme leader mondial dans le domaine du développement préclinique d'agents anti-arthrosiques, tout en permettant au GREPAQ de la Faculté de médecine vétérinaire - Université de Montréal d'avancer dans la compréhension de cette maladie chez l'animal de compagnie, chat ou chien. Au final, les répercussions pour le bien-être animal amélioreront la qualité de vie des animaux et de leurs propriétaires.
