The overall goal of this proposal is the set-up of a high-throughput synthetic biology and bioprocess optimization platform to rapidly synthesize genes and create gene and strain libraries, and use them for applications ranging from 1) metabolic engineering, 2) enzyme discovery via metagenome screening, and 3) protein engineering. As part of this grant, we would like to establish a high-throughput robotic system to enable a workflow starting from synthesized DNA, plasmid library creation, strain construction, strain selection from colonies, enzymatic and growth assays for characterization of strains and the proteins and finally, bioprocess optimization and scale-up studies.
