Chemtrade produces an extensive line of highly versatile aluminum-based Vaccine Adjuvants (AVA) that can be used over a broad range of pH, types of protein binding, and other vaccine requirements. The control of the processing parameters in the production of AVA control their physical properties, such as stability, rheology, particle size, which have an impact on their efficacy as vaccine adjuvants. The proposed research will focus on elucidating the relationship between the surface characteristics of AVA on their stability and efficacy as vaccine adjuvants. The key properties that will be quantified are, (a) particle size, (b) zeta potential, (c) morphology, (d) rheology and (e) surface composition of the nanoparticles. We will develop synthetic protocols to elucidate the parameters that control the colloidal stability of the Rehydragel adjuvants. Additionally, we will conduct QA analyses of the Rehydragel samples from production batches in order to elucidate the relationship between processing parameters and the physical properties of the AVA. The key objective of the project is to develop fundamental understanding on the colloidal chemistry of the AVA, and to use this knowledge to advance and improve the performance of the AVA, in terms of their long-term stability and their efficacy as vaccine adjuvants. Additionally, we will train highly qualified personnels to support the development and production of better AVA to support the growing demands and needs of vaccine in human and veterinary applications.
