对反应性星形胶质增生及瘢痕形成调控机制的研究是脊髓损伤研究领域内的重要课题。本项目基于课题组的前期研究,发现Ski在大鼠脊髓损伤后活化的星形胶质细胞中高表达,沉默星形胶质细胞中Ski基因的表达后,其增殖及迁移能力均显著降低。我们初步提出Ski可能是一种新的调控脊髓损伤后反应性星形胶质增生及瘢痕生成的重要分子的科学假说。为进一步证实,我们将采用基因沉默和过表达技术研究大鼠脊髓损伤后Ski对反应性星形胶质增生、胶质瘢痕形成及神经功能恢复的影响。另外,本项目还将对Ski可能通过Ras-Raf1-ERK1/2通路调控大鼠脊髓损伤后反应性星形胶质增生及瘢痕生成的机制作初步研究。通过本项目的研究,我们将首次明确Ski在脊髓损伤后反应性星形胶质增生及瘢痕生成过程中的调控作用,为脊髓损伤的病理机制研究及治疗提供新的方向。
英文摘要
The regulation mechanism of reactive astrogliosis and glial scar formation after spinal cord injury(SCI) is an important topic in the research field of SCI. This project is based on the previous research of our group. we found that Ski was highly expressed in activated astrocytes after SCI in rats, and its proliferation and migration ability were significantly reduced after the expression of Ski gene was silenced in astrocytes. Therefore, we propose a scientific hypothesis that Ski may be a novel molecule that regulates reactive astrogliosis and glial scar formation after SCI. For further confirmation, we will use gene silencing and overexpression techniques to study the effect of Ski on reactive astrogliosis, glial scar formation and neural functional recovery after SCI in rats. In addition, this research will also make a preliminary study on the mechanism by which Ski may regulate reactive astrogliosis and glial scar formation after SCI in rats through Ras-Raf1-ERK1/2 pathway. From this study, we will define the regulatory role of Ski in the process of reactive astrogliosis and glial scar formation after SCI for the first time, and provide a new direction for the study on the pathological mechanism and treatment of SCI.
