青光眼患者人数随人口老龄化逐年攀升。眼压(IOP)升高是青光眼重要危险因素,促进房水外流是降低IOP的有效方法。青光眼患者小梁网外流通道受损后,葡萄膜淋巴外流通道有很好的潜在开发适用性,此途径将是青光眼药物治疗的新靶点,但如何促进其外流还有待研究。前期研究通过建立房水葡萄膜淋巴外流光声成像检测平台,首次发现此途径外流量随年龄增长而明显下降。可能与老龄后淋巴内皮功能下降,内皮型/诱导型一氧化氮合酶(eNOS/iNOS)活性逆转导致的NO利用度降低有关。大麻素受体激活后可以改善这种逆转,同时可以促进肠道淋巴运输。因此,我们推测眼内大麻素受体激活,可以改善老龄淋巴内皮功能,提高NO利用度,调控眼部淋巴管运动。本项目基于前期建立的光声成像检测平台,通过单次小鼠腹腔注射大麻素及其受体、通路因子拮抗剂,探索外源性大麻素对房水葡萄膜淋巴外流的影响,并揭示其所依赖的分子机制,为青光眼提供新的药物治疗策略。

The number of glaucoma patients continues to increase with age per year. Elevated intraocular pressure (IOP) is an important risk factor for glaucoma and promoting aqueous humor outflow is an effective method to reduce IOP. Uveolymphatic outflow pathway has potential development applicability when trabecular outflow pathway is impaired in glaucoma patients which will be a novel target for glaucoma drug treatment, but how to promote its outflow remains to be studied. In our previous research, for the first time, we found that the uveolymphatic outflow decreased significantly with age through the established photoacoustic imaging system platform. It may be related to the decline of lymphatic endothelial function and the decrease of NO utilization induced by endothelial nitric oxide synthase/inducible nitric oxide synthase (eNOS/iNOS) reversion with age. Activation of Cannabinoid receptor can ameliorate this reversal, and also promote intestinal lymph transport. Therefore, we hypothesize that the activation of cannabinoid receptor in the eye can improve the function of aged lymphatic endothelium, promote the release of NO and regulate the contraction of lymphatic vessels. This study aims to investigate the effects of exogenous cannabinoid on uveolymphatic outflow of aqueous humor and the associated molecular mechanisms in mice by single intraperitoneal injection of cannabinoid, its receptor and antagonists, in order to provide new drug therapeutic strategies for glaucoma.
