Juvenile hormone (JH) is crucial for the growth and silk protein synthesis of the silkworm, Bombyx mori. However, little is known about the regulation of juvenile hormones biosynthesis in the silkworm. We found that the molting and pupation of silkworm occurred in advance after the interference of POU-M2. Immuno-histochemical analysis revealed that POU-M2 was expressed in the corpora allata (CA) of the silkworm. RT-PCR analysis showed that the expression profiles of POU-M2 in the CA of silkworm were consistent with the trend of JH titer in the hemocyte. The expressions of the key enzymes in the JH biosynthesis signal pathway and Kr-h1 were up-regulated after the overexpression of POU-M2, and down-regulated after the interference of POU-M2. Dual luciferase assays showed that POU-M2 promoted the transcription of juvenile hormone acid methyltransferase (JHAMT) promoter. Therefore, we suggested that POU-M2 regulates the transcription of JHAMT in the corpora allata of the silkworm, thus regulating juvenile hormones biosynthesis and the development of silkworm. Studying the molecular mechanism of juvenile hormone biosynthesis regulated by POU-M2 in the silkworm not only facilitates to better understand the regulatory mechanism of the silkworm and lepidopteran insects development, but also could provide new molecular targets and theoretical guidance for the biological control of agricultural and forestry pests.
