提高卵母细胞发育潜能与成熟率,是改善女性生育力的核心问题之一。lncRNA可参与调控卵泡发育,抑制颗粒细胞凋亡,但其对卵母细胞成熟的作用和调控机制尚不清楚。lnc-ODNR是申请人在卵母细胞转录组数据中筛选到一条人鼠同源的lncRNA,其表达随卵母细胞成熟过程逐渐上调,沉默后可抑制Igf2r表达,降低卵母细胞成熟率。生物信息分析发现lnc-ODNR和Igf2r启动子区部分序列互补配对,且具有转录因子Sox30结合位点。故提出假说:lnc-ODNR招募Sox30激活Igf2r转录,促进卵母细胞成熟。本项目拟:①在体外水平探讨lnc-ODNR在卵母细胞成熟中的作用;②在体内水平揭示lnc-ODNR对卵母细胞成熟的影响;③在分子水平阐明lnc-ODNR募集Sox30特异性转录Igf2r的机制。本研究对揭示卵母细胞成熟机制有重要意义,可为提高卵泡发育潜能和改善女性生育力提供新线索。

How to improve development potential and maturation rate of oocytes is one of the core issues in improving female fertility. At present, the regulatory mechanisms of follicular development and oocytes maturation are poorly understood, the study of transcriptional regulation during oocyte maturation is of great significance for the treatment of ovarian function and fertility. A human mouse homologous lncRNA (lnc-ODNR) was selected from oocyte transcriptome database, and its expression was gradually up-regulated with oocyte maturation. Silencing of lnc-ODNR can decreased the maturation rate of oocytes. We found that some sequences of lnc-ODNR and Igf2r promoter region were complementary and paired, and there were potential sequences of Sox30 binding sites in lnc-ODNR. The silencing of Sox30 could also down-regulate the expression of Igf2r. lnc-ODNR was bound to Sox30 (proved by RIP experiment). Therefore, we proposed that: lnc-ODNR recruited Sox30 to activate Igf2r transcription, thus promoting oocyte maturation. The purpose of this project is to: ①investigate the role of lnc-ODNR in oocyte maturation in vitro; ②reveal the function of lnc-ODNR on follicular development in vivo; ③elucidate the molecular mechanism of lnc-ODNR recruiting Sox30 to activate transcription of Igf2r. This study will provide clues for new methods to improve oocyte maturation rate and female fertility.
