排卵是指成熟卵子从滤泡中释放出来的动态过程,受到复杂而精准的调控。前列腺素(Prostaglandin,PG)是调控排卵的关键因子,其很难自由通过生物膜,需要依赖前列腺素转运载体的运输。然而,有关前列腺素转运载体在哺乳动物排卵中的功能研究只有零星报道,在鱼类排卵中的研究更是一片空白。我们初步研究发现,前列腺素转运载体中负责PG外流的MRP4(Multidrug Resistance Protein 4 )与负责PG摄入的PGT (Prostaglandin Transporter)可能参与调控斑马鱼的排卵活动。鉴于此,本项目拟以斑马鱼为模型,选择MRP4与PGT为切入点,采用抑制剂处理、基因敲除及辅以PG补偿的方法,研究前列腺素转运载体对斑马鱼排卵过程中滤泡细胞内外PG浓度的调控以及对排卵的影响。本项目的研究将揭示前列腺素转运载体在鱼类排卵中的功能,也为其他脊椎动物的排卵调控研究提供参考。
英文摘要
Ovulation is a complex and precise biological process during which mature oocytes are released from the surrounding follicle wall into the ovarian cavity. It is well established that the PGs is one of the key fact during the ovulation. While it is noteworthy that PGs are diffuse poorly across model biological membranes and need relies on prostaglandin transporter. However, it have sporadic relate study about the function of prostaglandin transporter on ovulation in mammals. And even no study about the function of prostaglandin transporter on ovulation in fish. Our preliminary data suggest that MRP4 (responsible for PGs efflux) and PGT (responsible for PGs influx) are involved in regulate ovulation in zebrafish. Therefore, in this project, using zebrafish as the experimental model, two important prostaglandin transporters, including MRP4and PGT are selected,utilize inhibitor treatment, gene knockout and PGs replacement to investigate the roles of prostaglandin transporter on regulate the concentration between extracellular and intracellular and regulate the ovulation. The implementation of this project will reveal the function of MRP4 and PGT on ovulation in fish, and lead us to a better understanding of the regulation of ovulation in vertebrate.
生殖是物种得以繁衍的基础,对动物生殖活动及其调控机理的研究一直是人们最感兴趣的课题之一。物种在长期的进化过程当中,形成了精密的生殖调控策略,从而确保物种的正常繁衍。其中,排卵是指成熟卵子从滤泡中释放出来的动态过程,受到复杂而精准的生殖策略的调控。我们前期的研究表明,前列腺素(Prostaglandin,PG)参与排卵过程,并且发挥重要作用。然而,前列腺素很难自由通过生物膜,需要依赖前列腺素转运载体的运输。截止目前,有关前列腺素转运载体在哺乳动物排卵中的功能研究只有一篇报道,在鱼类排卵中的研究更是一片空白。鉴于此,本项目拟以斑马鱼为模型,以MRP4 (Multidrug Resistance Protein 4,负责PG外流)与PGT (Prostaglandin Transporter,负责PG摄入)作为切入点,采用抑制剂处理、基因敲除的方法,研究前列腺素转运载体对斑马鱼排卵过程中滤泡细胞内外PG浓度的调控以及对排卵的影响。本项目通过抑制剂处理发现PGT 抑制剂DIDS 显著降低卵泡内的前列腺素浓度,而MRP4 的抑制剂AEBSF 则显著上调卵泡内的前列腺素浓度。同时,通过将斑马鱼pgt 与mrp 4 基因敲除,发现具有类似的结果。更为重要的是,无论上调或者下调卵泡中的前列腺素的浓度,均会导致斑马鱼排卵受到一定程度的抑制。在此基础之上,我们进一步研究了前列腺素合成酶在斑马鱼排卵过程中的功能以及其他内固醇激素,包括雌激素和雄激素对斑马鱼生殖调控的研究。相关研究成果,我们作为5篇第一标注的SCI学术论文在本研究领域公开发表,待发表论文2篇。这些结果表明,卵泡内前列腺素浓度的平衡对于维持斑马鱼正常排卵具有重要的作用,相关研究成果也是对雌性生殖调控网络的重要补充。在该项目的资助之下,参加国际学术会议2次,国内学术会议3次,同时,协助培养研究生4名。项目投入经费26.00万元,支出25.8730万元,各项支出与预算基本相符。
