苏云金芽胞杆菌(Bt)是广泛使用的一种生防制剂,主要成分为杀虫晶体蛋白(Cry)和芽胞。许多Cry蛋白的表达依赖于芽胞形成,芽胞形成调控与Cry蛋白表达密切相关。关于Bt表达与调控的研究主要集中于cry基因,而对芽胞形成调控研究较少,芽胞形成调控机制仍待深入。前期发现一个转录活性很高、功能未知的deoR基因,可以显著影响Bt芽胞形成。在此基础上,本项目拟以Bt HD73菌株为研究对象,利用基因敲除、功能互补、基因过表达、点突变等功能基因组学、生化检测等方法,同时结合CHIP-seq及转录组学,明确DeoR的功能,揭示DeoR调控芽胞形成的分子机制。本项目预期结果不仅有利于解析Bt芽胞形成的分子调控机制,而且为Bt工程菌株改造和工业化生产奠定基础,进一步为Bt的田间应用提供科学依据。
英文摘要
Bacillus thuringiensis(Bt) is a kind of widely-used biocontrol agent with the main components of the mixture of insecticidal crystal proteins(Cry) and spores. Many Cry protein expression is closely connected to sporulation regulation since that the Cry proteins are expressed by a sporulation-dependent manner. However, the studies on genes expression and regulation in Bt strain mostly focused on those encoding Cry proteins, rather than the sporulation regulation. Thus, the regulatory mechanisms of sporulation in Bt still need to be further explored and verified. Our previous study revealed that a deoR gene with unknown function retained a very high transcriptional activity in a Bt strain based on the transcriptomics analysis, and deoR significantly affected sporulation. Based on these results, this project aims to reveal the function of DeoR and the molecular mechanisms of sporulation regulated by DeoR in Bt by combining functional genomics techniques, such as gene knockout, complementation, overexpression, site-directed mutagenesis and CHIP seq, transcriptomics analysis. Our finding will not only help reveal the molecular mechanisms of sporulation in Bt, but also lay the foundation for engineered Bt strains construction and industrial production, and further provide theoretical support for Bt application in the field.
苏云金芽胞杆菌(Bt)是一类广泛使用的生防菌,其杀虫活性依赖于Cry蛋白的表达,而芽胞形成与Cry蛋白表达密切相关。Bt HD73菌株可以毒杀鳞翅目害虫,对其Cry1Ac蛋白的调控研究较多,但其芽胞形成研究较少。本研究主要利用基因敲除、功能互补、基因过表达等功能基因组学方法,同时结合生化检测等,明确sirR基因在调控Bt HD73菌株中对芽胞形成、Cry1Ac蛋白的表达及母细胞裂解的调控作用。前期分析转录组数据发现,sirR基因 (HD73_5014) 转录水平很高。本研究分析SirR蛋白序列,表明其仅含一个helix-turn-helix (HTH) 保守性结构域,预测为DNA结合蛋白,在芽胞杆菌属中保守性较高;该基因缺失后加速不对称隔膜的形成,并且母细胞裂解的时间提前;通过lacZ融合菌株的构建和β-半乳糖苷酶活性的测定,证明在细胞生长前期,∆sirR菌株中Spo0A~P水平较野生型中的高,表明sirR通过调控Spo0A~P的活性影响细胞进入芽胞形成阶段;sirR基因的缺失影响芽胞形成进程然而不影响最终的芽胞形成率和Cry蛋白产量;∆sirR突变体中,cwlC基因的转录时间提前,进而促进母细胞裂解的提前;纯化SirR蛋白,经测定,SirR蛋白无体外碱性磷酸酶活性;利用GFP标记sirR基因,通过荧光显微镜观察发现SirR蛋白均匀分布于母细胞的细胞质中,进一步证实了SirR为转录调控因子。本研究明确了sirR基因在Bt芽胞形成和母细胞裂解中的调控作用。研究结果将为有效解决Bt生产实际应用问题提供科学依据,并且为新一代Bt制剂的分子设计开辟遗传改良的新途径。
