中医认为肾藏精、主骨、生髓,骨骼的退变和肾气的衰退紧密相连。在细胞及分子层面,肾藏精主要体现在干细胞及其调节的微环境。胚肾细胞能够发育成多种肾脏固有细胞,具有干细胞属性,即肾精的属性,但能否促进成骨细胞成骨分化,尚不清楚。本研究以中医经典理论为指导,①通过已经申请专利的提取方法,分离、纯化小鼠胚肾细胞,并鉴定其多向分化潜能;②利用细胞共培养体系,通过检测成骨分化标志物,明确胚肾细胞单独及联合补肾强骨中药有效成分柚皮苷促进成骨细胞成骨分化的作用;③通过条件共培养、细胞因子芯片及中和抗体等技术,筛选候选功能蛋白,并通过正向、负向干预,揭示胚肾细胞促进成骨细胞成骨分化的分子机制;④体内观察胚肾细胞单独以及联合柚皮苷对小鼠低转运型肾性骨病骨转运状态的影响,并提供形态、结构和分子生物学证据。旨在进一步发掘肾主骨中医藏象理论新的科学内涵,为低转运型肾性骨病提供新的治疗思路。
英文摘要
The traditional Chinese medicine (TCM) believes that the kidneys store the essence, control the bone and produce the marrow. The regression of bone closely related to the decline of renal function. At the cellular and molecular level, it mainly reflected in the stem cells and the niche it regulated. The embryonic renal cell (ERC) contains renal stem cells, which can differentiate into the permanent renal cells. However, it remains unclear whether they are able to promote the osteogenesis of osteoblasts. On the basis of classical TCM theory, ①we plan to use a patent of our research group, to extract and purify ERCs, and to testify their multiple differentiation potentials; ②using the co-culture system, to explore the effect of ERCs on the osteogenesis of osteoblast with or without the exposure of naringin;③using cytokine microarrays to investigate the effects of cytokines secreted by the ERCs on the osteogenesis of osteoblasts in vivo and to find out the effective cytokines and pathways; ④ to investigate the effectiveness of the ERCs and naringin on the bone of low turnover renal osteodystrophy and to collect the morphological, pathological and molecular evidence. This study aims to enrich and develop scientific connotations of the theory of visceral manifestations and to provide new clues for the treatment of low turnover renal osteodystrophy.
