P-选择素及其配体影响ACC-M细胞肺转移的实验研究

P-选择素通过与其相应配体结合而介导肿瘤细胞与宿主细胞间粘附反应，进而参与恶性肿瘤的转移过程。以P-选择素免疫双缺陷鼠为模型，证实P-选择素在ACC-M细胞肺转移过程中起丶饔玫幕∩希掷搿⒋炕疨-选择素配体蛋白;制备相应的单抗。验证该抗配体单抗对ACC-M细胞肺转移的阻断作用。加深对恶性肿瘤转移机理的认识，为癌转移的防治提供新思路?.

Metastasis is a major reason for the death of malignant tumor patients. To investigate the effect of P-selectin and its ligand on the metastasis of oral cancer, we have completed many research with Institute of Biochemistry and Cell Biology, Chinese academy of Science. The conclusion is that some new P-selectin ligands have been detected on human tongue cancer Tca8113 cells, malignant melanoma A375 cells, salivary gland carcinoma ACC-M cells and breast cancer ER-75-30 cells. They are heparan-like proteoglycans ligand and glycoprotein ligand. These ligands may play a critical role in mediating adhesion of human tumor cells. In vitro interrupting P-selectin binding to the ligand can reduce the adhesion of tumor cells and palatelets. In vivo monoantibody and heparan can inhibit ACC-M cells adhesion to the lung of nude mice and reduce the incidence of lung metastasis to 96.30% and 92.75%. 6 papers have been finished. 2 of them were published on 《The Journal of Immunology》. The conclusion is that 1) P-selectin and the ligands mediate the metastasis of oral tumor, 2)heparan and P-selectin monoantibody have an good treating effect to oral gland carcinoma.
