在冠心病的发病及死亡人群中,老年冠心病患者具有年龄相关性静息高交感神经活性的特点,交感神经活性与氧化应激密切相关,降低氧化应激被认为是改善老年患者冠状动脉内皮细胞功能的治疗方式之一,但尚无有效可行的抗氧化方式。本项目组前期研究发现,去肾交感神经(RDN)可以有效降低机体氧化应激水平,但未见RDN在治疗老年冠心病方面的研究。我们提出假说:RDN通过降低氧化应激水平改善老年患者冠状动脉内皮细胞功能。线粒体是衰老、氧化应激、内皮细胞相互联系的关键细胞器,本项目拟将高龄动脉粥样硬化小鼠作为研究对象,以内皮细胞线粒体作为靶器官,探讨:1.RDN能否通过降低氧化应激水平改善冠状动脉内皮细胞功能;2.线粒体在RDN对冠状动脉内皮细胞功能影响中的作用机制。验证RDN通过降低氧化应激提高线粒体功能改善内皮细胞稳态,为老年冠心病患者心血管疾病的防治提供新的思路。

Elderly patients account for more than half of the morbidity and mortality of coronary heart disease.Elderly patients with coronary heart disease have the characteristics of age-related high sympathetic nerve activity at rest, and sympathetic nerve activity is closely related to oxidative stress. Reducing oxidative stress is considered as one of the treatment methods to improve the function of coronary endothelial cells in elderly patients, but no effective and feasible antioxidant methods have been found.Previous studies of our team have found that Renal Sympathetic Denervation (RDN) can effectively reduce the oxidative stress level, and there has been no research on the treatment of elderly coronary heart disease by RDN.We proposed the hypothesis that RDN could improve the function of coronary endothelial cells in elderly patients by reducing the level of oxidative stress.Mitochondria are the key organelles for aging, oxidative stress and endothelial cell interaction. Therefore, this project intends to take elderly atherosclerotic rats as subjects and mitochondria of endothelial cells as the target organ to explore: 1. Whether RDN can improve the function of coronary endothelial cells by reducing the oxidative stress; 2. The role and mechanism of mitochondria in the improvement of RDN on the function of endothelial cells.It will verified that RDN could improve endothelial cell homeostasis by reducing oxidative stress and improving mitochondrial function, providing a new idea for the prevention and treatment of cardiovascular disease in elderly patients with coronary heart disease.
