光合作用是影响油菜产量的关键因素,叶色突变体是研究光能利用率的理想材料。本项目前期研究通过EMS诱变,获得了一个可稳定遗传的叶色黄化突变体,其叶绿素含量及光合速率与野生型相比显著降低。遗传研究表明,该表型由1对隐性基因控制,并将控制叶色黄化的基因BnA03.chd定位于A03染色体上814Kb的区间内。在此基础上,本项目拟对BnA03.chd进一步精细定位,通过比对已公布的甘蓝型油菜基因组序列,结合拟南芥同源基因的功能,确定目的基因。通过互补实验及基因敲除对目的基因进行功能验证,并进行表达模式分析及亚细胞定位,解析目的基因引起叶色黄化的分子机理,为深入研究光合作用影响因素,提高油菜光合速率提供理论基础。
英文摘要
Photosynthesis is the key factor affecting the yield of rapeseed, and the leaf color mutant is an ideal material to study photosynthetic efficiency. A heritable yellow-leaf mutant was obtained by EMS mutagenesis. The chlorophyll content and photosynthetic rate of the mutant were significantly lower than those of the wild type. Previous studies confirmed that the phenotype was controlled by a recessive gene BnA03.chd, which was located in an 814 Kb interval on chromosome A03. On the basis of these results, in this project we intends to make further fine mapping of BnA03.chd, and get the target gene through the comparison of the published genome sequence of Brassica napus, combine with the function of Arabidopsis thaliana homologous genes. The transgenic verification of the target gene will be carried out by complementary experiment and gene knockout, and the expression pattern analysis and subcellular localization will be carried out to analyze the molecular mechanism of yellow-leaf caused by the target gene. The result will provide a theoretical basis for further study on the factors influencing the photosynthesis, and genetic improvement of photosynthetic rate in rapeseed.
