低血钙是围产期奶畜的频发代谢病,对奶业生产造成严重影响。解析低DCAD提高血钙水平、防治低血钙的作用机理是学界持续致力解决的重要科学课题。基于低DCAD能正向调控钙特异入胞与胞内转运的研究结果,项目假设:低DCAD可上调小肠PMCA表达水平,易化钙跨膜外排出胞入血,提高血钙水平,防治低血钙。为此,项目以围产期奶山羊为试验动物,从“整体-组织-细胞”3个水平层面,应用细胞分子生物学技术手段结合经典动物饲养试验,开展以下研究内容:①从整体水平上把握低DCAD可提高血钙含量。②从组织水平上验证低DCAD可提高小肠PMCA表达水平,同时可促进钙跨膜外排。③从细胞水平上验证低DCAD介导PMCA调控胞内钙跨膜外排入血。项目创新点:①逻辑推论性强,学术思路新颖;②研究系统性强,是前期研究内容的拓展与延伸。完成上述研究内容,可验证项目假说,完整揭示低DCAD防治低血钙的作用机理,保障奶山羊持续稳产。
英文摘要
Hypocalcemia is a frequently occurred metabolic disease in transition dairy animals and results in a dramatic loss in dairy industry. Reducing dietary cation-anion difference (DCAD) can increase blood calcium level and then prevent hypocalcemia. However, the mechanism behind it remains unclear and is still a focused research field for nutritionist. Based on the positive result that decreasing DCAD is able to regulate Ca2+ inflow into cell and transportation, it is hypothesized that reducing DCAD could enhance PMCA expression level in small intestine to facilitate dietary Ca2+ extrusion into blood for improvement of blood Ca2+ level, and ultimately, prevent hypocalcemia.. In order to test our hypothesis, 3 research contents will be conducted with transition dairy goats served as experimental animals from animal herd-tissue-cell levels. During the process, modern molecular techniques and classical animal feeding trial will be adopted together. The 1st content is conducted to evaluate the effect of lowering DCAD on blood Ca2+ concentration increase from animal herd level. The purpose of the 2nd content is to testify that, from the tissue level, reducing DCAD can increase PMCA expression level in small intestine and the promote Ca2+ extrusion across transcellular. Content 3 is to examine that reducing DCAD is involved in mediating Ca2+ outflow into blood through the existence of PMCA from the cellular level. . Two innovations is manifested in the present project. The first one is the academical innovation due to the logical inference. The second one is the strong systematicness because it is the extension based on our previous research. . The completion of the above-mentioned research contents will be able to test our hypothesis and clarify the mechanism why reducing DCAD is effective to prevent hypocalcemia. It can also lay a scientific basis for the sustainable production of dairy goats in the practice.
