鱼粉替代是对虾营养研究的热点,负责人前期研究发现植物蛋白替代鱼粉较多时容易诱发炎症反应和内质网应激,同时代谢组学结果表明:鱼粉水平降低后,在饲料DHA水平一致的前提下,DHA及含有DHA的磷脂是凡纳滨对虾血淋巴中下降最显著的代谢物。DHA发挥抗炎作用的核心机制是通过改变生物膜磷脂组成影响细胞免疫信号传导。因此提出科学假说:对虾体内DHA水平的降低导致内质网膜不饱和度降低、通透性提高,Ca2+从内质网释放,从而诱发内质网应激并诱导细胞凋亡。本项目首先通过蛋白印迹、免疫组化和Ca2+检测明确Ca2+稳态在高植物蛋白诱导内质网应激产生和发展过程中的作用,进而研究DHA是否通过提高内质网膜不饱和度抑制Ca2+紊乱和内质网应激。同时构建Ca2+紊乱诱发的对虾原代肝胰腺细胞内质网应激模型,验证DHA是否通过调节Ca2+稳态缓解内质网应激。本项目预期结果将为低鱼粉水平下保障对虾健康养殖奠定重要基础。

Fishmeal replacement is the hot field of shrimp nutrition research, our previous study found inflammation and endoplasmic reticulum stress (ERS) would be induced when fishmeal was replaced by high plant proteins, metabolomics results indicated that after fishmeal was replaced, although DHA levels were similar among treatments, DHA and DHA contained phospholipid were the most significant decreased metabolites in hemolymph of Litopenaeus vannamei. DHA plays its anti-inflammatory role in affecting of the immune response signaling through changing the membrane phospholipid composition. So we put forward a scientific hypothesis: the decrease of DHA decreased the unsaturated degree and permeability of ER, and release Ca2+ from ER, finally inducing ERS. This project is firstly conducted to evaluate the role of Ca2+ homeostasis in the emergence and development of high plant protein induced ERS through the Western blot, immunohistochemistry and Ca2+ detection. Secondly, we would investigate whether DHA have the ability to inhibit ERS and Ca2+ disorder through increasing the unsaturated degree of ER membrane. Then Ca2+ disorder induced primary hepatopancreas ERS model of shrimp would be built to investigate whether DHA inhibit ERS through regulating the Ca2+ homeostasis. The prospective result of this project would lay the foundation for health culture of shrimp fed low fish meal diet.
