新型冠状病毒肺炎疫情过后,SARS-CoV-2对人类健康的远期影响将是亟待探讨的新课题。女性卵巢上也有SARS-CoV-2受体——血管紧张素转化酶2(ACE2),因此SARS-CoV-2感染对女性生殖健康的影响不容忽视。研究证实:(1)已知的冠状病毒感染使ACE2下调;(2)ACE2是肾素-血管紧张素系统(RAS)重要的负调节因子,ACE2下调将导致RAS功能紊乱;(3)卵巢RAS功能紊乱与多种生殖功能异常有关。我们推测既往SARS-CoV-2感染使ACE2表达下调,继而使卵巢RAS功能紊乱,导致女性生殖功能异常。本课题拟:(1)分析既往SARS-CoV-2感染对女性生殖功能的影响。(2)探讨既往SARS-CoV-2感染对颗粒细胞和卵母细胞ACE2表达水平的影响。(3)利用ACE2基因敲除小鼠模型,验证上述影响和机制。本研究将有助于我们预测未来人们生育能力的变化,制定应对措施,造福社会。
英文摘要
After the epidemic of the new coronavirus pneumonia, the long-term effects of SARS-CoV-2 on human health will be new research that needs to be explored. It was reported that angiotensin-converting enzyme 2 (ACE2) is used by SARS-CoV-2 for entry into human cells. Further, ACE2 expression has been identified in the human ovary and is a member of the renin-angiotensin system (RAS), which has a central role in the female reproduction system. It was reported that some human coronavirus infections reduce ACE2 expression. Thus, it may be hypothesized that a decrease in ACE2 expression following SARS-CoV-2 infection may disturb ovarian RAS, causing reproductive disorders. There will be three aspects of the research in this study. First, we will explore the impact of previous SARS-CoV-2 infection on female reproductive function. Second, we will evaluate the association between the previous SARS-CoV-2 infection and the level of ACE2 expression in granulosa cells and oocytes. Finally, the effect and mechanism of previous SARS-CoV-2 infection on female reproductive function will be verified by an ACE2 gene knockout mouse model. This research will help us predict future changes in people's reproductive function and take countermeasures to benefit our society.
