This work has studied the preventive and therapeutic effect of vasonatrin peptide (VNP) on the hypoxic right ventricular hypertrophy(HRVH).Experiments in vivo suggested that VNP could significantly decrease right ventricular index ,average pulmonary artery pressure and pulmonary vascular resistance of HRVH rats. Effect of VNP was stronger than that of C-type natriuretic peptide(CNP) and atrial natriuretic peptide(ANP). Cell culture experiments suggested that VNP could inhibit the proliferation and collagen secretion of cardiac fibroblasts. VNP could also inhibit the hypertrophy of cardiac myocytes induced by moderate hypoxia. VNP had no effect on the survival rate of severe hypoxic cells but reduced the activity of lactate dehydrogenase. Molecular biology experiments suggested that VNP could reduce the level of NPR-C and ET-1 in hypoxic rat hearts and cultured cardiac myocytes. In conclusion , VNP is a strong anti-proliferation factor which can prevent and cure HRVH.
