为高效防范疯牛病、适应饲料质量安全监管发展需求,本研究提出肉骨粉种属鉴别的新技术路线。采用LC-MS和GC-MS深入挖掘不同种属肉骨粉及反刍动物源成分掺加样品的特异性蛋白和脂质组成;采用MALDI-TOF-MS确定肉骨粉种属鉴别的特异性蛋白和脂质的高标示性特征信息;解析特异性蛋白和脂质在MALDI-TOF-MS上的表征与信息贡献,建立肉骨粉种属鉴别分析方法与模型,探究检测机理;建立基于MALDI-TOF-MS的肉骨粉的特异性蛋白和脂质的定量分析模型;揭示特异性蛋白和脂质质谱信息的有机融合与互补机制,建立基于两组分多特征谱段的肉骨粉种属鉴别的精准识别模型;建立基于特异性蛋白和脂质的MALDI-TOF-MS的反刍动物源成分掺加样本的定性鉴别及定量分析模型。该方法有望与国际动物源性饲料标准方法进行有效联用,对于保障动物源性饲料监管措施和促进饲料产业的可持续发展,具有重要的科学意义和实际应用价。
英文摘要
This study focuses on the discrimination of different animal-derived meat and bone meal (MBM) in order to effectively prevent BSE and develop feed quality and safety supervision. A new idea and technical route are proposed based on a large number of reliable samples and previous research. The Liquid Chromatography-mass spectrometry (LC-MS) and gas chromatography-mass spectrometry (GC-MS) are used to further explore the composition and structure of specific proteins and lipids of different MBMs and animal-derived feeds adulterated with ruminant ingredients. The characteristics and differences of mass spectral information of specific proteins and lipids of MBMs are studied by matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF-MS), and the information contributions of specific proteins and lipid on MALDI-TOF-MS spectra are analyzed to establish a scientific and reliable discrimination analysis method and to provide theoretical basis to clarify the detection mechanism. The quantitative analysis of specific proteins and lipids of different MBMs is explored by MALDI-TOF-MS. The combination and complementary mechanism based on the mass spectrometry information of specific proteins and lipids are analyzed to establish a precise identification model based on two-component multi-characteristic spectra for MBMs. The qualitative and quantitative analysis model of animal-derived feeds adulterated with ruminant ingredients is established based on specific proteins and lipids using MALDI-TOF-MS. This method is expected to be effectively combined with the current international standard method for the detection of animal-derived feeds, which have important scientific significance and practical application value on the effective guarantee for the control of animal-derived feed and the promotion of stable and sustainable development of the feed industry.
