人工晶状体植入术是矫正白内障摘除后无晶体眼的最有效方法,但术后炎症引起蛋白和细胞在人工晶状体上粘附,导致其透明度下降影响手术效果。申请人前期发现,聚二甲基硅氧烷分子刷在玻璃表面接枝可形成一种低表面能的类液涂层,能显著提高疏水性和抗生物粘附性能。据此我们首次制备了具有类液涂层的人工晶状体,并初步证明其具有高疏水性和优异的生物相容性,既能抗蛋白和细胞粘附又有望与后囊膜紧密贴附,且具备一定的自清洁特性。本项目将进一步通过人工晶体检查仪器和抗生物粘附实验全面评价类液涂层人工晶状体的光学特征和自清洁功能,阐释其抗生物粘附的自清洁机制;其次,建立人工晶状体动物植入模型,通过光学性能、病理学和细胞分子生物学检测,系统研究类液涂层人工晶状体下调眼内炎症的效能与机制,并动态追踪其植入后生物相容性及愈合转归,有望深入阐释人工晶状体的生物相容机制,开发可降低炎症等相关并发症和光学性能长期稳定的人工晶状体。
英文摘要
Intraocular lens implantation is the most effective method to correct aphakia with high refractive error after the cataract extraction. However, postoperative inflammation induces adhesion of proteins and cells to intraocular lens, which leads to the decline of optical properties and influences the effect of operation. In our previous study, polydimethylsiloxane molecular brush grafted on glass surface could form a new liquid-like coating with low surface energy, which could significantly improve the hydrophobicity and self-cleaning function with anti-biological adhesion ability. According to our previous research, we have created the intraocular lens with liquid-like coating for the first time, and preliminarily proved it has high hydrophobicity and excellent biocompatibility, which can not only resist the adhesion of proteins and cells, but also has great potential to attach tightly with the posterior capsule with self-cleaning properties. This project will further evaluate the optical characteristics and self-cleaning function by the intraocular lens testing instruments and comprehensive anti-biological adhesion experiments to explain the self-cleaning mechanism of anti-biological adhesion. Additionally, in the animal model of intraocular lens implantation, we will systematically study the efficiency and mechanism of the intraocular lens with liquid-like coating in lowering intraocular inflammation, by the detection of optical properties, pathology and cell molecular biology. Furthermore, the biocompatibility and healing outcome after implantation were dynamically evaluated. We aim to thoroughly illustrate the biocompatibility mechanism of intraocular lens and develop a new intraocular lens which can reduce inflammation and its related complications with long-term optical property.
