英文摘要
For mammals, the successful reproduction relies largely on the quality of an embryo. The sows are in negative energy balance after birth, which can cause reduced quantity and quality of eggs, and results in decline of the litter size and lifetime reproduction.Melatonin (MT) can improve the quality of female eggs, and increase the litter size and lifetime reproductive for female animals. MT promotes egg maturation and embryonic development in a variety of animals through its receptor or antioxidant effects. It has been found that MT can promote the maturation of pig eggs in vitro and induce modifications on fatty acid metabolism. The preliminary research shows that MT can improve the litter size and affect the type of fatty acids in pig eggs, but the specific regulatory mechanism is undetermined. We proposed that MT regulated the synthesis of fatty acids and its oxidation in mitochondria to improve egg quality and developmental competency. To this end, the effect of MT on fatty acids-oxidation and the synthesis ATP by mitochondria in pig eggs was studied first by methods using live cell ATP probes.Secondly, the effects of MT regulation of pig egg lipid synthesis and peroxide were explored by combining live cell staining and lipotomic assays, And finally, the effects of MT regulation of fatty acids beta-oxidation and peroxidation in pig eggs on their maturation and developmental competency. This project is expected to reveal the mechanism of melatonin function in modulating fatty acid metabolism to promote the maturation of pig eggs, and could provide an insight for improving the reproductive efficiency and life time reproductivity of sows.
