骨骼肌损伤临床患病率高,常因反复损伤、衰老、代谢失调而导致骨骼肌纤维化或萎缩,易引发慢性疼痛和再次损伤,严重影响运动和降低生活质量,为亟待解决之临床难题。本项目组既往国自然项目已证实推拿手法能有效抑制骨骼肌损伤后纤维化,而本课题系该方向上的进一步延伸,综合生物力学、细胞生物学、分子生物学研究方法,以期立足于推拿手法的机械力特征,基于机械敏感离子通道Piezo,序贯探讨推拿手法促进骨骼肌损伤后再生的机制,验证“手法可通过Piezo蛋白通道引起Ca2+离子内流,激发Ca2+/PI3K/Akt信号通路,从而调控巨噬细胞极化与肌卫星细胞的转化增殖,进而导致成肌细胞增殖,最终促进骨骼肌损伤再生修复”的假说。本项目探索推拿生物效应的启动机制,探讨手法促进骨骼肌修复的准确靶点,为深入认识推拿手法生物力学激发机制提供新的实验依据,有助于推拿疗法在临床上更广泛、更精准的应用,对促进推拿学科发展具有重要意义。
英文摘要
The clinical morbidity of skeletal muscle injury is high. It often causing fibrosis or atrophy of skeletal muscle due to repeated injury, aging, metabolic disorders, and easily cause chronic pain and re-injury, seriously affect exercise and reduce the quality of life, which is a clinical problem to be solved urgently. The Past National Natural projects of this project group have proved that massage manipulation can effectively inhibit fibrosis after skeletal muscle injury. This study is a further extension of this direction. Comprehensive research methods of biomechanics, cell biology and molecular biology. Because of the mechanical characteristics of massage manipulation, based on the mechanical Piezo sensitive ion channels, discuss sequential massage promote regeneration mechanism, after injury of skeletal muscle validation "technique can be caused by Piezo protein channel flow within the Ca2 + ions, stimulate the Ca2 + / PI3K/Akt signal pathway, so as to regulate macrophage polarization and satellite cell proliferation,Finally promote the hypothesis of "skeletal muscle injury regeneration and repair".At the same time, the initiation mechanism of the biological effect of massage was explored, and the accurate target of the manipulation of massage to promote skeletal muscle repair was explored, providing a new experimental basis for the in-depth understanding of the biomechanical excitation mechanism of manipulation of massage, which is conducive to the more extensive and more accurate application of massage therapy in clinical practice, and is of great significance for promoting the development of the discipline of massage.
