根系限制Na+向地上部转运是葫芦科作物耐盐的普遍方式,前期研究发现盐胁迫下印度南瓜(Cucurbita. maxima)比中国南瓜(Cucurbita. moschata)叶片中积累更多盐害离子Na+,却表现出更强耐盐能力,表明葫芦科作物中存在根系限制Na+向地上部转运以外的耐盐途径。利用2份叶片Na+积累差异显著材料进行叶脉转录组测序,筛选到位于南瓜叶脉中一个Na+/H+反转运蛋白CmNHX4,结合X-Ray能谱分析初步推测该蛋白通过向叶脉皮层细胞区域化Na+而降低盐害离子对叶片的伤害。以此为基础,本项目拟将印度南瓜CmNHX4转化中国南瓜,通过基因编辑技术敲除印度南瓜CmNHX4,验证其基因功能,并利用H2O2生成抑制剂DPI研究ROS在调控CmNHX4功能中的作用。本课题完成对阐明印度南瓜组织耐盐具有重要意义,同时为葫芦科耐盐育种提供全新思路。

Limiting Na+ transport to the shoot by root is a common way to resist salt stress in Cucurbitaceae crops. However, in our previous research work, we have found that under the salt stress, the Cucurbita. maxima accumulated more toxic level of Na+ in the leaves than in the Cucurbita. moschata, but possesses higher salt tolerance, indicating a novel salt tolerance strategy in Cucurbitaceae crops besides the Na+ restriction strategy in roots. In the prior experiments, we found that this shoot-based salt tolerance mainly relies on the Na+ regionalization in the leaf veins of Cucurbita. maxima controlled by a Na+/H+ anti-transport CmNHX4 (Based on the RNA-seq with two distinguished materials). X-Ray EDS show that CmNHX4 was preliminarily presumed to be localized Na+ to the vein cortex cells. And reduce the damage of salt harmful ions to the photosynthetic system. Based on the results, this project aims to analyze the function of CmNHX4 in Cucurbitaceae leaves, by suppressing or overexpressing CmNHX4 in Cucurbita. maxima and Cucurbita. moschata. And use the H2O2 production inhibitor DPI to study the role of ROS in regulating the function of CmNHX4.The results may be of important theoretical significance for salt tolerance breeding of Cucurbitaceae.
