利用代谢工程优化特定代谢物的生物合成是调控植物次生代谢的重要手段。丹酚酸B是丹参质量控制指标之一。我们前期发现,水杨酸引发的细胞质酸化促进了丹参丹酚酸B的积累,但不清楚该信号如何调控丹酚酸B生物合成。本项目拟从调控胞质pH的质膜H+-ATPase入手,对响应水杨酸诱导的丹参质膜H+-ATPase编码基因进行筛选与分析,利用基因过表达、沉默等基因工程技术解析质膜H+-ATPase对丹参丹酚酸B生物合成的调控功能。本研究对阐明质膜H+-ATPase调控植物次生代谢的作用机制具有重要意义,为完善水杨酸引发的胞质酸化信号转导对丹参次生代谢物的调控机制提供重要的理论依据。

It has been demonstrated that the metabolic engineering approaches could optimize the biosynthesis of specific metabolites. Salvianolic acid B is one of the quality control indicators in Salvia miltiorrhiza. It was found that salicylic acid-induced cytosolic acidification promoted the accumulation of salvianolic acid B in our previous work. Unfortunately, it is unclear that how the cytosolic acidification regulate the biosynthesis of salvianolic acid B. Therefore, we start this research from plasma membrane (PM) H+-ATPase, which has functions to regulate cytosolic pH homeostasis. The aim of this proposal is to reveal the regulating function of salvianolic acid B synthesis by PM H+-ATPase using gene engineering technology with gene silencing and overexpression. The results could not only be significance in illunimating the regulating mechanism in plant secondary metabolism by PM H+-ATPase, but also provide the theoretical basis for signal transduction pathway of secondary metabolism in Salvia miltiorrhiza.
