营养不亲和与食用菌的杂交育种、品系分类和病害传播研究密切相关,子囊菌的营养不亲和多数情况下由het位点调控。目前梯棱羊肚菌栽培发展迅猛,新品系选育和病害防控研究迫在眉睫。该项目用前期获得的遗传背景清楚且来自同一子囊果的梯棱羊肚菌单孢菌株YPL6-3(MAT1-1型)和YPL6-1(MAT1-2型)混和播种,从产生的一个子囊果中挑取数个子囊,分离其中的8个成熟孢子作对应标记并鉴定交配型,通过这些单孢菌株群体间及与两个亲本菌株间的营养不亲和性鉴定,关联它们的het和mat位点基因序列结构、表达水平差异,探寻这些菌株间营养不亲和产生是否与子囊来源相关,是否由het和mat位点调控;通过具有互补交配型的营养亲和及不亲和菌株对的出菇实验,明确营养亲和性对子囊果生长发育的影响。本项目研究工作可以探索梯棱羊肚菌营养不亲和产生的分子调控机制,为梯棱羊肚菌杂交育种和病害防控研究提供理论基础。
英文摘要
Vegetative incompatibility (VI) has to be considered when studies on cross breeding, strain classification and pathogen transmission of the edible mushroom are carried out. In Ascomycota fungi, the VI is controled by genetic differences between the two individuals at het loci. In recent years, with the rapid expanding of the cultivation area of Morchella importuna, it is extremely urgent to carry out the studies on cross breeding and disease control. In previous study, we had acquired the M. importuna single ascospore strains YPL6-3(MAT1-1) and YPL 6-1 (MAT1-2) which isolated from a ascocarp YPL6, and analyzed their genome information. In this study, after sowing the mixed YPL6-3 and YPL6-1, one ascocarp will be selected to pick several asci and isolate 8 ascospores within every ascus, then all the single ascospore isolates will be identified mating type and marked one to one correspondence. The VI reactions between these sibling isolates and also between their parental strains will be tested. At the same time, for these strains, the het and mat loci genes cloning and expressing will be preformed. After correlation analyses between VI reactions and genetic data, we will attempt to explore that whether the VI of M. importuna is related with the ascus and mediated by the het or mat loci. Some cultivation experiments of the vegetative incompatibility pairs and the vegetative compatibility pairs will be conducted, in order to answer whether VI influences the fruiting bodies forming or not. These works will reveal the molecular mechanisms that mediated the Vegetative incompatibility of M. importuna, and will contribute to studies on cross breeding and disease control.
