中药质量控制是保证其临床用药安全性和有效性的重要措施,也是实现中药现代化与国际化的关键科学问题。中药质量标志物(Q-markers)是其物质基础的集中体现,故有必要建立基于Q-markers的质量评控方法。本项目在前期研究基础上,提出“中空纤维双功能”的细胞/靶标捕获与液相微萃取相结合的方法,对中药Q-markers进行体内外筛选、辨析及测定,从而制定特征性的中药质控策略。利用分别固定细胞和/或靶标以及充满有机溶剂的中空纤维,在莪术、苦参和半枝莲药材及其饮片的体内外样品中直接筛选。通过化学、生物学技术交叉融合,联通中药固有和其体内效应成分之间关系,辨析Q-markers并对其定量;揭示Q-markers和中药有效性的关联度、药材-饮片质量的溯源性以及多成分、多靶点的药效机制,开展包含Q-markers的(半)定量测定研究,为构建符合中药特色的特征性质控方法,提供新思路和简便易行的操作平台。

Quality control of traditional Chinese medicine (TCM) is an effective measure to ensure the safety and effectiveness of TCM in clinical practice, which is also one of the critical scientific issues in realizing its modernization and globalization. Quality markers (Q-markers) of TCM are representative of material basis. Therefore, it is necessary to develop a characteristic quality control method based on Q-markers for TCM. Based on previous research, a strategy of characteristically evaluating quality of TCM is proposed by our group by combining hollow fiber cell/target fishing method with hollow fiber liquid phase microextraction. In this research, different cells and targets are seeded or loaded in the lumen and inner wall of hollow fiber, both which and the hollow fiber impregnated with organic solvent are inserted into extract or biologically sample of TCM to capture, discriminate and determine bioactive constituents and Q-markers of curcumae rhizoma, sophorae flavescentis radix, scutellariae barbatae herba and their decoction pieces. The correlation of bioactive Q-markers in vitro (herbs) and in vivo (biological samples), the relevance between Q-markers and efficacy of TCM, and the traceability of quality between decoction pieces and TCM herbs will be probed and elucidated. The therapeutic mechanism of “multicomponent and multitarget” of TCM will be studied. Furthermore, Q-marker-based fingerprint and multicomponent determination method will be established. In summary, the project can provide new ideas and a simple operating platform to discriminate Q-markers, and develop a quality control method meeting the characteristics of TCM.
