基于细胞自噬mTOR信号通路调控研究益气温阳护卫法防治支气管哮喘的作用机制

哮喘患者细胞自噬水平明显升高，气道上皮细胞是哮喘发生的初始部位，在哮喘的发生、发展过程中有着重要作用。抑制上皮细胞自噬，可以改善哮喘症状。嗜酸性粒细胞自噬在严重哮喘病理过程中发挥重要作用，是哮喘慢性持续气道炎症的重要原因之一。中医“卫气”与自噬存在一定的相关性，可通过提高卫气功能来调节自噬从而提高机体的抗病能力。以往的研究已证实益气温阳护卫法具有降低气道反应性，明显延长哮喘发作的潜伏期，并通过调节Th1/Th2细胞分化实现。但是否进一步通过mTOR信号通路调控细胞自噬实现防治哮喘的目的，尚未明确。由此，本课题基于细胞自噬mTOR信号通路调控研究益气温阳护卫法防治支气管哮喘的作用机制。课题组拟采用体内、体外实验，通过RT-PCR 、Western blot检测、MDC染色，荧光显微镜检测等方法研究益气温阳护卫法对细胞自噬及相关mTOR信号通路的影响。为益气温阳护卫法防治哮喘提供科学依据。

The level of autophagy in asthma patients increased significantly, and airway epithelial cells were the initial site of asthma, which played an important role in the occurrence and development of asthma. The inhibition of epithelial autophagy can improve asthma symptoms. Eosinophilic autophagy plays an important role in the pathological process of severe asthma and is one of the important reasons for chronic continuous airway inflammation of asthma. Chinese medicine "Wei Qi" has a certain correlation with autophagy, and can improve the body's ability to resist disease by improving the function of Wei Qi. Previous studies have confirmed that Yiqi Wenyanghuwei method has reduced airway responsiveness, significantly extended the incubation period for asthma attacks, and achieved by regulating Th1/Th2 cell differentiation. However, it is not yet clear whether the purpose of controlling cell autophagy through mTOR signal pathway will be further achieved. Therefore, this topic studies the mechanism of prevention and treatment of bronchial asthma based on cell autophagy mTOR signal pathway regulation. The task group intends to use in vivo and in vitro experiments to study the effects of the gas warming method on cell autophagy and related mTOR signal pathways through RT-PCR, Western blot detection, MDC staining, and fluorescence microscope detection. It provides scientific basis for the prevention and treatment of asthma.
