嗅觉对昆虫的生存至关重要。对昆虫嗅觉感受机理的深入研究是探索害虫专一性防治的基础。目前,对雌性性信息素的嗅觉感受机理研究颇多,但对雄性性信息素的了解却很少。疆夜蛾是黄淮流域新发生的有潜在爆发为害能力的重要农业害虫,该虫雄蛾释放的唯一性信息素组分为2-苯乙醇。我们前期的研究结果表明2-苯乙醇可引起疆夜蛾雌雄成虫的触角电位反应,并在疆夜蛾两性交配中发挥重要作用。本项目将首先利用爪蟾卵母细胞表达/双电极电压钳记录技术从疆夜蛾触角转录组中鉴定2-苯乙醇的气味受体基因,之后采用CRISPR/Cas9基因编辑技术并结合电生理检测以及行为学试验,进一步验证并明确该受体基因的功能。研究结果有望填补雄性性信息素嗅觉感受机理研究的匮乏,对完善蛾类昆虫的性信息素通讯机制具有重要意义,并且可为其它昆虫雄性性信息素受体基因的鉴定提供参考,同时也可为寻找新的害虫生物防治途径提供思路和理论依据。
英文摘要
Olfaction is essential for insect survival. In order to control the insect pests more efficiently and selectively, it is necessary to explore and understand the mechanism of their olfaction. At present, there are many studies on the olfactory mechanism of female sex pheromones, but little understanding of male sex pheromones. Peridroma saucia Hübner is an emerging agricultural pest that has the potential to cause severe outbreak in Huanghuai River Basin. 2-Phenylethanol was identified as the sole component of male sex pheromone of P. saucia. Our previous studies showed that 2-phenylethanol could induce electroantennogram responses in male and female adults and play an important role in their mating. In this project, we will first identify the odorant receptor of 2-phenylethanol from the antenna transcriptome with Xenopus oocyte expression and two-electrode voltage-clamp recording. Then, we will adopt CRISPR/Cas9 gene editing method combined with electrophysiology and behavioral tests to further verify and clarify the function of this receptor. The results are expected to fill in the lack of research on the olfactory mechanism of male sex pheromone, which is of great significance to supplement the olfactory mechanism of sex pheromone in moths, and can provide reference for the identification of odorant receptor of male sex pheromone in other insects. Moreover, it can also provide ideas and theoretical basis for finding new ways of biological control of pests.
