脱氧核糖核酸(DNA)和生物硫醇是与人类遗传信息和重大疾病密切相关的活性分子。要阐明DNA和生物硫醇的生理功能和病理机制的相互关系,需要建立一个在生物体内对DNA附近生物硫醇可视化的动态监测方法。然而,现有的DNA或生物硫醇探针的研究忽略了DNA和生物硫醇在活体内的相互依存关系,无法实现对生物体内DNA附近生物硫醇的动态监测。针对这一关键科学问题,探针的研究需要构建新型的肿瘤靶向性近红外DNA/生物硫醇荧光探针。本项目首先预发展新型的高稳定性DNA近红外荧光探针,以DNA探针为平台引入肿瘤靶向位点和生物硫醇识别位点,进一步构建肿瘤靶向性近红外DNA/生物硫醇荧光探针。该探针有望为解决目前所面临的无法动态监测癌细胞和肿瘤内DNA附近生物硫醇的关键科学问题提供理论指导和实验依据,具有重大的科学意义和应用价值。
英文摘要
Deoxyribonucleic acid (DNA) and biological thiols are active molecules closely related to human genetic information and major diseases. To clarify the relationship between physiological function and pathological mechanism of DNA and biological thiols, a dynamic monitoring method visualizing biological thiols near DNA in organisms needs to be established. However, the existing studies of DNA or biological thiols probes ignore the interdependence between DNA and biological thiols in biological systems, so that the dynamic monitoring of biological thiols near DNA cannot be realized. So, to meet the urgent need of current biomedical research, it is important to build tumor-targetable DNA/biological thiols fluorescent probe with good photostability. A novel high photostability DNA near-infrared fluorescence probes are pre-constructed in this project. Based on the above probe, the tumor target sites and biological thiols recognition sites were introduced to these structures to develop novel DNA near-infrared fluorescent probes with high photostability. These probes can achieve dynamically monitoring of the biological thiols near DNA in cancer cells and tumors. This project aims to solve issue that the near infrared fluorescence probe is unable to dynamically monitor the biological thiols near DNA in cancer cells and tumors, which provides theoretical guidance and experimental basis, and has great scientific significance and application value.
