芽胞杆菌脂肽fengycin对尖孢镰刀菌具有很强的拮抗作用,能够开发为新型稳定的生物农药。脂肽的抗真菌活性与结构密切相关,其能够通过诱导真菌凋亡方式实现其抗菌作用,然而fengycin对尖孢镰刀菌的构效关系和凋亡机制还未解析。前期研究发现,地衣芽胞杆菌FJAT-4产生的fengycin有6个不同单组分组成,诱导尖孢镰刀菌呈现凋亡特征。本项目将利用多种技术进行FJAT-4 fengycin不同组分的纯化和结构表征,分析脂肽结构与抗真菌活性的关系;检测fengycin诱导尖孢镰刀菌细胞凋亡时活性氧累积、caspase激活、线粒体膜电位、钙离子及细胞色素C含量变化等;结合蛋白质组和转录组筛选差异蛋白和基因,通过功能分析确定凋亡关键基因,挖掘凋亡靶点,揭示fengycin诱导尖孢镰刀菌凋亡作用机制,同时评估脂肽对瓜类枯萎病的生物防治效果。研究结果为脂肽在植物真菌性病害生物防治中的开发应用奠定基础。
英文摘要
The lipopeptide fengycin has strong antagonistic effect on Fusarium oxysporum and can be developed as a new stable biopesticide. Studies have shown that the antifungal activity of lipopeptides is closely related to structure, and it exerts antifungal effects by inducing apoptosis-like death. However, the structure-activity relationship and apoptosis mechanism of fengycin against F. oxysporum are not clear. Our earlier studies found that the fengycin produced by Bacillus licheniformis FJAT-4 is composed of six different components, which induces the apoptosis of F. oxysporum. This project is aim to determine the structure and conformation of fengcyin and to analyze the relationship between lipopeptide structure and antifungal activity; to detect the accumulation of reactive oxygen species, caspase activation, Mitochondrial membrane potential, intracellular Ca 2+ and cytochrome C levels of F. oxysporum induced by fengycin ; to identify the key apoptosis-related genes and to find apoptotic target of F. oxysporum induced by fengycin using proteome and transcriptome; to evaluate the biological control efficiency of lipopeptide on cucurbit wilt. Above research is of great significance for the development and the application of lipopeptide in the biocontrol of plant fungal diseases.
