乳香是治疗糖尿病血管病变的重要中药。前期研究发现乳香醋炙后降糖和活血作用显著增强,体现炮制独特优势,但增效机制有待深入。已证实,脂肪酸代谢与糖尿病血管病变密切相关,而蛋白IRF2BP2可激活VEGFA保护血管,调控脂肪酸代谢改善胰岛素抵抗。初步研究表明,乳香醋炙后及主要变化成分ADHBA能促进IRF2BP2高表达。因此提出假说:乳香醋炙后可能通过增强IRF2BP2蛋白表达,调控机体脂肪酸代谢,增强糖尿病血管病变改善作用。.本研究通过糖尿病模型大鼠明确醋炙对乳香的增效作用;采用代谢组学技术探究乳香醋炙前后模型大鼠脂肪酸代谢差异;从mRNA和蛋白水平,揭示乳香醋炙前后及ADHBA对IRF2BP2及脂肪酸代谢相关分子表达的影响;利用CRISPR/cas9技术,验证IRF2BP2对乳香醋炙调控脂肪酸代谢环节作用的机制,从而阐明醋炙增强乳香改善糖尿病血管病变的机理,对于丰富传统炮制理论具有重要意义。

Frankincense is of significance in the treatment of diabetic angiopathies. According to the guide of traditional processing theory, our research group conducted a series study proving the enhancement efficacy of vinegar-processed frankincense in decreasing blood glucose levels and promoting blood circulation. It reflects the unique advantages of traditional Chinese medicine processing. However, the mechanisms that underpin the efficacy enhancement of processing are not fully understood. Fatty acid metabolism is closely related to diabetic vascular disease. And IRF2BP2 was reported to activate VEGFA protecting blood vessels and regulate fatty acid metabolism improving insulin resistance. Vinegar-processed frankincense augmented the expression of IFR2BP2 was proved preliminarily. Hence the hypothesis is that vinegar-processed frankincense up-expresses IRF2BP2 and regulates the metabolism of the fatty acids, thereby enhancing the improvement of diabetic angiopathies..In this project, the diabetic model rats were applied to investigate the synergistic effect of vinegar-processing; metabolomics was used to explore the changes in fatty acid metabolism in diabetic rats administrating frankincense and vinegar-processed frankincense; conducting molecular biology techniques to reveal the effects of frankincense, vinegar-processed frankincense and ADHBA on the expression of IRF2BP2 and fatty acid metabolism-related molecules; Using CRISPR/cas9 gene editing technology to verify the mechanism of IRF2BP2’s key role in regulating fatty acid metabolism of frankincense vinegar-processing. This project is from the perspective of the regulation of fatty acid metabolism mediated by the key protein IRF2BP2 to reveal the improving effect of vinegar-processed frankincense on diabetic angiopathies. And it provides scientific support for the traditional processing theory.
