系统性红斑狼疮(systemic lupus erythematosus,SLE)是一种累及机体多系统、多器官的自身免疫性疾病。其发病机制尚不明确,前期研究证实解毒祛瘀滋肾方对系统性红斑狼疮确切的临床疗效,研究同时发现补肾类中药具有调节间充质干细胞的功能特性。而间充质干细胞(mesenchymal stem cells,MSCs)是一类中胚层来源的非造血干细胞,是造血微环境的重要成分,具有免疫调节及诱导免疫耐受能力、免疫原性弱、组织修复再造等特点。本项目研究结合前期研究结果,继而从分子和蛋白水平入手,探讨解毒祛瘀滋肾方促进衰老MSC形态学、增殖分化能力、移植修复能力的改变及与Wnt/β-catenin信号通路相关性分析等方面,进一步客观地阐述中药调节间充质干细胞的功能特性与SLE之间的内在相关性,为解毒祛瘀滋肾方治疗系统性红斑狼疮提供有力的临床指导。

Systemic lupus erythematosus (SLE) is an autoimmune disease involving multiple systems and organs. Its pathogenesis is not clear. Previous studies confirmed the exact clinical effect of Jiedu Quyu Zishen formula on systemic lupus erythematosus. At the same time, it was found that kidney tonifying herbs have the function of regulating mesenchymal stem cells. Mesenchymal stem cells (MSCs) are a kind of mesoderm derived non hematopoietic stem cells, which are important components of hematopoietic microenvironment. They have the characteristics of immune regulation and induction of immune tolerance, weak immunogenicity, tissue repair and reconstruction. Based on the results of previous studies, this project, starting from the molecular and protein levels, explored the effects of Jiedu Quyu Zishen Recipe on the morphology, proliferation and differentiation of aging MSC In order to provide a strong clinical guidance for the treatment of systemic lupus erythematosus, the change of transplantation and repair ability and the correlation analysis with Wnt / β - Catenin signal pathway were further objectively elaborated between the functional characteristics of mesenchymal stem cells regulated by traditional Chinese medicine and SLE.
