无膜细胞器是细胞内某些蛋白质或者核酸发生相分离而形成的液滴。已有的研究表明相分离在细胞中广泛存在,相分离的失调通常与一些神经退行性疾病相关。对于相分离形成的分子机制以及相分离过程中蛋白质构象的变化的研究对理解蛋白质的聚集机制以及由聚集所导致疾病的发病机理至关重要。SNAP25蛋白是人体内广泛分布的蛋白质,我们发现SNAP25蛋白在体外会发生相分离。目前对SNAP25发生相分离的分子机制仍不清楚。本项目拟运用常规以及19F核磁共振波谱方法研究SNAP25发生相分离过程中SNAP25结构变化、相互作用信息以及相分离液滴中SH3蛋白的稳定性,理解SNAP25发生相分离的分子机制,为SNAP25形成相分离的生物学功能研究打下基础。19F NMR研究相分离具有广泛的应用性,能推广到其他类型的相分离系统(蛋白质、RNA)研究中。

Membraneless organelles are droplets formed by the phase separations of certain proteins or nucleic acids inside cells. Previous studies have revealed that phase separations exist extensively in cells. Dysfunctions of phase separations are often associated with some neurodegenerative diseases. The study on molecular mechanisms of phase separation and protein conformational changes during phase separation are critical for understanding the mechanism of protein aggregation and the pathogenesis of diseases caused by aggregation. The SNAP25 protein is a widely distributed protein in humans. We found that SNAP25 protein underwent phase separation in vitro. The molecular mechanism of phase separation of SNAP25 remains unclear. This project intends to use conventional and 19F NMR spectroscopy methods to study the structural changes and protein interactions of SNAP25 and probe SH3 stability in phase-separated droplets during phase separation of SNAP25. We seek to study conformational changes and protein interactions at the atomic level and explore the role of these changes and interaction in phase separation process. We hope this study would contribute to the molecular mechanism understanding of SNAP25 phase separation and lay a foundation for the study on the biological function of SNAP25 phase separation. 19F NMR studies on phase separation have a wide range of applications and will be applied to the study of other phase separation systems (protein and RNA).
液液相分离现象常见于各种生物过程中,在转录调节、翻译调节、DNA缩合、mRNA加工、RNA稳定性、核糖体蛋白组装、细胞增殖和分裂中都具有重要作用。液液相分离液滴的环境高度拥挤,各种相互作用广泛存在,这些环境因素对处于液液相分离内蛋白质的稳定性的影响目前尚不清楚。基于此,我们运用核磁共振技术研究蛋白质及核酸在液液相分离中的稳定性及构象交换信息。首先探索DDX4N1形成相分离的条件,运用相分离液滴实现了对蛋白质和G-四链体的有效包裹。运用19F NMR技术研究了SH3在DDX4N1相分离液滴中、高浓度的DDX4N114FtoA溶液中及大分子拥挤环境中的热力学稳定性,探索相分离影响SH3蛋白稳定性的分子机制。结果表明在BSA及DDX4N114FtoA溶液,弱相互作用占主导作用,使SH3稳定性显著降低,未形成相分离时仅能观测无结构态的峰。相分离液滴会降低SH3稳定性,其局部有序结构可以减弱与蛋白质的弱相互作用,相比于DDX4N114FtoA溶液,可以提高蛋白质的稳定性,并且其疏水表面的暴露相比稀溶液中有所减小。运用19F-19F exchange研究了相分离中SH3的交换动力学,结果显示相分离液滴中在ms-s时间尺度内未观测到SH3的构象交换信息,并且交换速率的改变是由于相分离液滴中的局部有序结构所引起的。运用CMPG RD实验研究了GB3-MutX在相分离液滴中的构象分布及构象交换速率,结果表明GB3-MutX在相分离液滴中构象分布未发生变化,但是构象交换速率下降了50%。运用19F NMR技术研究了G4结构在相分离液滴中的构象变化,结果显示,G4结构在相分离液滴中消失了,而相分离液滴被蛋白酶K破坏后G4结构又重新恢复,说明G4结构不是被降解而是发生了解链。存在G4时,相分离液滴不能够被蛋白酶K降解,G4对DDX4N1的某些片段具有保护作用。我们的结果显示相分离液滴可以改变包裹其中的生物大分子的物理化学性质,相分离液滴所形成的局部有序结构及不均一性微环境对于调节生物分子的性质至关重要。
