牙鲆弹状病毒(HIRRV)是一种传染性强,致死率高的鱼类病毒,近年来在亚欧地区广泛流行,已成为我国鱼类健康养殖的重要威胁。流行病学调查显示HIRRV的发生与流行与水温密切相关,但目前有关温度影响HIRRV侵染及鱼体抗病毒应答特征研究仍处于空白。本项目在前期病原分离、流行特点调研、单抗研制及靶组织/细胞鉴定等基础上,解析不同温度下HIRRV基因在牙鲆及细胞系中的转录与表达差异特征,并利用研制的抗HIRRV单抗追踪病毒在关键靶组织/细胞中的侵染差异;同时,利用研制的抗牙鲆T/B淋巴细胞亚群特异性抗体,监测不同温度下各亚群淋巴细胞差异响应特征,进而在分选获得T/B淋巴细胞群的基础上利用组学技术系统解析病毒关键靶组织/细胞对HIRRV侵染的差异应答特征;同时检测不同温度下牙鲆关键抗病毒相关基因及特异性抗体差异应答水平;以期深入揭示温度影响HIRRV感牙鲆染的细胞与分子机制,为疾病防控提供理论基础。

Hirame novirhabdovirus (HIRRV) is a kind of rhabdovirus with strong infectivity and high lethality. It was widely spread in Asia and Europe in recent years, which has been becoming a vital threat of marine fish aquaculture. Epidemiological survey revealed that the outbreak of HIRRV was closely related with the water temperature, but research on how temperature influence the infection of HIRRV and the antiviral response of fish is almost blank. Based on our previous work, the present project aims to analyze the differences in transcription and expression of early genes of HIRRV in flounder and cell lines (HINAE) at different temperatures, and track infection process and proliferation dynamic of HIRRV in the key target tissues and cells using the developed anti-HIRRV monoclonal antibody; On the other hand, the differential response characteristics of lymphocytes in each subpopulation at different temperatures were monitored by using the specific antibodies against flounder T/B lymphocyte subsets. Moreover, based on the T/B cell subpopulations obtained by sorting, the differential responses to HIRRV infection in key target tissues (spleen) / cells (lymphocytes) were analyzed by using the transcriptomics technology; Meanwhile, the key anti-virus related genes, non-specific immune response and antibody response were also detected. We aim to reveal the molecular mechanism of the influence of temperature on infection process of HIRRV and host anti-viral response, and provide the important theoretical basis for HIRRV prevention and control.
牙鲆弹状病毒(HIRRV)是一种传染性强,致死率高的鱼类病毒,近年来在亚欧地区广泛流行,已成为我国鱼类健康养殖的重要威胁。流行病学调查显示HIRRV的发生与流行与水温密切相关,为了解析温度影响HIRRV侵染及鱼体抗病毒应答特征,项目在阐明了牙鲆弹状病毒CNPo2015株全基因组序列特征,研制出抗HIRRV-G蛋白单克隆抗体,进而利用多组学技术系统全面解析了HIRRV重要靶器官——牙鲆脾脏在不同温度条件下的抗病毒应答特征,发现HIRRV感染能够显著活化牙鲆RIG-I样受体信号通路,且该通路在牙鲆适温条件下(20℃)的响应水平与响应速度显著强于低温易感条件(10℃);阐明了升温与降温调控对HIRRV增殖及牙鲆抗病毒免疫应答的影响特征,发现升温调控可有效激活CD8+ T淋巴细胞响应,并阐明了在温控条件下HIRRV感染可有效诱导牙鲆免疫保护性响应,并成功建立HIRRV活病毒免疫策略;发现HIRRV可侵入牙鲆T、B淋巴细胞但无法在胞内有效增殖,在对T/B淋巴细胞分选的基础上,阐明了不同温度条件下B淋巴细胞对HIRRV的差异应答特征,深入揭示了B淋巴细胞内BCL11A可通过与HIRRV基因组相互作用进而抑制病毒增殖的分子机制;另外,项目还利用研制的病毒单抗研发了HIRRV胶体金快速检测试纸,并研制出HIRRV乳酸菌活载体口服疫苗及HIRRV-G蛋白不同节段亚单位疫苗,其表现出良好的免疫保护效果。项目研究结果深入揭示了温度影响HIRRV感染牙鲆的细胞与分子机制,为疾病防控提供理论基础。
