木本植物一年多季开花现象,具有重要科研意义与应用价值。牡丹(Paeonia suffruticosa Andr.)是我国的传统名花,发展前景广阔,然而,花期短而集中,是长期制约产业发展的关键因素之一。本项目拟以具有连续开花性状的‘正午’牡丹为研究对象,通过与正常一次开花牡丹品种不同花期转录组序列差异分析,筛选并克隆磷脂酰乙醇胺结合蛋白(PEBP)基因家族中的抑花素基因(TFL1),进而通过转基因、基因沉默验证以及栽培措施验证等技术手段,及与成花素基因(FT)间蛋白互作与表达调控模式的分析,验证牡丹TFL1基因的功能以及该基因与‘正午’牡丹连续开花性状的关系,结合FT以全面阐释牡丹连续开花性状的分子调控机制,从而为牡丹花期改良育种提供科学基础与基因资源,并为木本观赏植物一年多季开花研究提供新资料。
英文摘要
Continuous blooming of woody plant species are very valuable both for the study and for the application. The tree peony (Paeonia suffruticosa Andr.) is the known-well traditional flower native to China, and, however, a concentrated and short flowering duration of it has become one of the key factors restricting the development of its industry. Using the special germplasm of tree peony hybrid ‘High Noon’ that could continuously flower several times in a growth season, this study will scan and clone the gene terminal flower 1 (TFL1) of Phosphatidylethanolamine-binding family protein (PEBP) gene family through a comparative analyses on the transcriptome sequences of different flowering-period between ‘High Noon’ and normal flowering tree peony cultivar. Transgenic test in Arabidopsis thaliana, gene silencing detection by VIGS and RNAi techniques, and culturation measure verification will be employed to identify the function of TFL1 gene. Gene expression and interaction of genes will be adopted to evaluate the relationship between TFL1 and FT genes in regulating continuous flowering of ‘high Noon’. The results would provide the scientific basis and genetic resources for the breeding to improve flowering in tree peony as well as the novel references on understanding the continuous flowering of other woody ornamental species.
