帕金森病(PD)是中老年人常见的神经系统变性疾病。已有的患者尸检病理数据显示,α-突触核蛋白是PD病变特征路易小体的主要成分。并有实验表明该蛋白及突变体会首先在多巴胺(DA)神经元的末梢异常聚集,而后逆行导致胞体的变性,即其轴突变性先于胞体变性。但如何导致该轴突变性的分子机制还尚未清楚。我们基于已有研究首次提出了α-突触核蛋白突变体可能通过干扰DA神经元轴突末梢局部蛋白的正常表达,而诱发了轴突变性的假说。为验证该假说本项目以已构建的α-突触核蛋白A53T过表达PD小鼠为基础,以核糖体免疫共沉淀(TRAP)技术为核心,拟通过高通量测序获得轴突局部mRNAs的表达图谱,继而应用生物信息学手段得到A53T组与对照组差异表达的基因及相关通路,来进一步了解α-突触核蛋白的突变体是如何通过改变轴突末梢局部蛋白的表达而诱发轴突变性的。本项目进一步探索了轴突变性机制,为PD病理成因及药物研发提供了新线索。

Parkinson’s disease (PD) is a chronic and progressive neurodegenerative disorder characterized by the loss of dopaminergic neurons in the substantia nigra and the presence of Lewy body. With the advent of global aging, the number of PD patients grows faster especially in China. α-Synuclein is the main component of Lewy bodies. Whereas both missense (such as A53T) and multiplication mutations of α-synuclein can cause the early-onset autosomal dominant familial form of PD. Previous studies indicated that the mice with expressing human A53T mutant showed α-synuclein aggregates at the presynapses of Dopaminergic (DA) neurons, where it physiologically located. Importantly, in some PD-like mouse models, such abnormal aggregation caused the axon degeneration first and soma degeneration later on. However the mechanism that underlines this interesting phenomenon is still unclear. Based on previous studies, we proposed that the overexpression of α-synuclein A53T may first disrupt the local translation of axon terminals in dopaminergic neurons, and lead to the axon degeneration afterwards. To address this hypothesis, a new technology TRAP was applied, through which mRNAs with high activity can be obtained. After comparing the mRNA expression profiles from axon terminals between control and A53T mouse model, the differentially expressed genes would be identified. To study these candidate genes and related pathways, we can better know how a-synuclein regulates the local translation of the axon terminals in DA neurons, which may be directly related with the axon degeneration.
本项目原定的计划是利用核糖体免疫共沉淀(TRAP)技术,拟通过高通量测序获得轴突局部mRNAs的表达图谱,来了解PD模型小鼠是如何通过改变轴突末梢局部蛋白的表达而诱发轴突变性的。但是在实验中遇到了三转基因模型小鼠(Pitx3-tTA/tetO-EGFP-L10a/tetO-A53T)的繁殖瓶颈(其一是三转基因小鼠本身获得的概率比较低,其二是大连新冠疫情的多次反复使得小鼠饲养、取材受到了很大的影响)。所以在项目开始的多半年时间里,整体进展十分缓慢,因此我们决定利用已成功构建的创新性的PD小鼠模型THCre/Pitx3fl/fl(Pitx3cKO)来研究多巴胺能神经元轴突末梢微环境在PD进程中的改变对其主要投射区—纹状体区SPNs神经元的影响。因此虽然所用的小鼠模型改变,但项目的原研究总旨还未改变,即探索PD的发展进程中多巴胺能神经元的轴突末梢的微环境是如何变化的。已知在PD中,纹状体经历进行性DA耗竭,导致主要运动症状,如静止性震颤、运动迟缓、姿势不稳定和僵硬。相应地,DA的主要突触后神经元SPNs也经历了形态生理的改变,为了更好地了解PD进程中纹状体神经元功能障碍的病理生理学,我们利用Pitx3cKO小鼠模型,该模型具有典型的PD特征,包括SNc DAergic神经元的显著丧失和运动缺陷。而且我们发现在衰老过程中,纹状体区的靶向神经递质、SPNs形态和DNA甲基化谱在Pitx3缺乏时发生显着改变。具体而言,在模型的早期纹状体中的多巴胺、GABA和谷氨酸在减少。而在老年的模型小鼠的SPNs中,细胞核、胞体和树突结构均出现萎缩以及细胞核内陷增加。此外,我们的结果显示在衰老过程中,SPNs的特征是出现更多的DNA损伤,而Pitx3缺乏加剧了这种现象,以及这种损伤的加剧同时伴随着与脂蛋白和核通路相关的DNA甲基化谱的改变。因此,SPNs内神经递质的早期扰动可能导致晚期纹状体内代谢、形态学和表观遗传学的改变,这可能为PD的诊断和发病机制的研究提供新的视角。
