妊娠高血糖致胚胎眼发育畸形的机理尚不清楚。研究发现高糖下氧化应激导致GTP氧化生成8-oxo-dGTP,提示妊娠高血糖致胚胎眼畸形可能与8-oxo-dGTP抑制眼发育调节枢纽因子Cdc42活化有关,但尚需科学验证。傣医认为 “四塔五蕴”即体内环境和物质代谢平衡是胚胎正常发育的关键,恢复氧化应激造成的内环境失衡可避免胚胎畸形发生。麻夯荒是傣族地区重要的药食两用植物。前期研究发现麻夯荒能修复高糖所致鸡胚眼畸形,缓解鸡胚眼内的氧化应激,促进Cdc42活化,并能降低H2O2负荷小鼠胚胎成纤维细胞中8-oxo-dGTP水平。综上,本项目拟在妊娠糖尿病小鼠胚胎和高糖鸡胚模型上研究麻夯荒改善胚胎眼畸形的作用及其作用机制,并筛选出麻夯荒中的活性化合物。本项目旨在阐明麻夯荒及其活性化合物抑制妊娠高血糖胚胎中8-oxo-dGTP促进Cdc42活化防治眼畸形发生的作用机制,为麻夯荒的应用提供科学依据。
英文摘要
The exact pathophysiology of embryonic eye malformation caused by gestational hyperglycemia remains poorly understood. Recent studies showed that the activation of Cdc42, one of the regulator hub during eye development, was impaired by 8-oxo-dGTP generated by oxidative stress due to the high glucose environment. This evidence indicates that the key role of inactive Cdc42 caused by 8-oxo-dGTP in the pathophysiology of eye malformation, which needs to be proved by scientific study. The theory of Dai medicine holds that the balance of “Si Ta Wu Yun” which means the inner environment and metabolism is critical to embryonic development. Rebalancing the oxidative stress of embryo can prevent embryonic malformation. In our preliminary research, Tamarindus Indica could attenuate the oxidative stress and restore the damage of chicken embryo eyes caused by high glucose and activate Cdc42. Tamarind could also lower the level of 8-oxo-dGTP in mouse embryonic fibroblast treated with H2O2. Taken together, we plan to study the inactive Cdc42 caused by 8-oxo-dGTP in pathophysiology of embryonic eye malformation resulted from gestational hyperglycemia. For such a purpose, we will determine the effects of tamarind on 8-oxo-dGTP under oxidative stress, and investigate how tamarind improves the dysfunctional Cdc42 by employing gestational diabetes mellitus mice model and chicken embryo model. We will screen out active compounds in tamarind. This project aims to clarify the pathophysiology of eye malformation caused by gestational hyperglycemia, reveal the protective effects of tamarind, and provide scientific evidences for the prevention and treatment of eye malformation caused by gestational hyperglycemia.
