褐飞虱是水稻的重要害虫,具有繁殖力高等特性,极易暴发成灾。研究表明长链非编码RNA(lncRNA)可能参与调控褐飞虱的繁殖力。前期我们通过对高繁殖力和低繁殖力基因型褐飞虱转录组的比较分析和大规模RNAi筛选,获得一个与胰岛素受体InR2共位置的候选lncRNA,暂命名为LncInR2。在此基础上,本项目拟对LncInR2进行全长克隆、组织和龄期表达特征分析、亚细胞定位及编码能力分析,明确其表达特征;对LncInR2进行RNA干扰敲低和CRISPR/Cas9敲除,分析其通过胰岛素信号通路对褐飞虱繁殖力的调控作用;最后,验证LncInR2与InR2的互作关系,并通过转录组测序鉴定其可能的竞争性miRNA,探索lncInR2调控褐飞虱繁殖力的分子机制。预期成果将阐明LncInR2通过胰岛素信号通路调控褐飞虱繁殖力的分子机制,为褐飞虱等农业害虫防控新策略和新技术提供理论依据。

The brown planthopper (BPH), Nilaparvata lugens, is one of the most important rice pest and is highly prone to outbreaks due to its high fecundity. It has been reported that long noncoding RNAs (lncRNA) participate in the regulation of BPH fecundity. In our preliminary results, we obtained a candidate lncRNA through the comparative transcriptomic analyses between the high fecundity genotype and low fecundity genotype of BPH and large-scale RNAi screening. The candidate lncRNA was designated as LncInR2 due to its co-location with InR2 which encodes an insulin receptor. On this basis, the following work will be carried out: 1) Clone the full-length sequence of LncInR2, and analyze its temporal and spatial expression pattern, coding probability, and subcelluar localization to understand its expression profile. 2) Knockdown and knockout LncInR2 by RNAi and CRISPR/Cas9, respectively, to analyze the regulation of lncInR2 on the fecundity of BPH by detecting the expression and phosphorylation of the key components in the insulin signaling pathway. 3) Identify the interaction between lncInR2 and InR2, and the potential competitive mRNA by RNA sequencing to explore the regulatory mechanism of LncInR2 on the fecundity of BPH. The results will reveal the molecular mechanism of lncInR2 in the regulation of BPH fecundity through insulin signaling pathway, and will provides theoretical basis for the new strategies and techniques to control BPH and other agricultural pests.
