“长脖芦”是传统鉴定中优质人参的关键品相特征之一,然而“长脖芦”的科学内涵尚未揭示。申请者前期发现人参干细胞内存在类似于拟南芥的CLE-WOX反馈调节环,由于拟南芥CLE-WOX反馈调节对植物根与茎的发育具有核心作用,由此推测人参的PgCLE-PgWOX反馈调控很可能对“长脖芦”形成具有关键意义。本项目提出:“长脖芦”的形成与根茎的茎尖干细胞及维管束干细胞最相关;人参根茎的茎尖及维管束干细胞命运由PgCLE与PgWOX反馈调控决定;基于PgCLE-PgWOX的反馈调控环研究人参芦头的发育可以有效解析“长脖芦”的形成机制。本项目以PgCLE与PgWOX反馈调控为突破口,开展人参芦头的结构发育研究,从分子生物学及细胞生物学方法揭示“长脖芦”的形成机制,为优质品相人参的选育提供理论基础。
英文摘要
“Long-necked rhizome” is one of the key characteristics of high quality ginseng by traditional identification, yet its scientific connotation has not been revealed. Applicants have previously found that ginseng has a feedback regulation loop similar to CLE-WOX of Arabidopsis thaliana, which determines the development of various organs of ginseng in an orderly manner to form different characteristics of ginseng. Thus, PgCLE and PgWOX feedback regulation loop is supposed to be closely related with the development of ginseng rhizomes. This project proposes that, the formation of “long-necked rhizome” is most related to the stem cells of shoot apical meristem and vascular meristem; the fate of the above stem cells is determined by PgCLE and PgWOX regulation. Based on the feedback regulation loop of PgCLE and PgWOX, studying the development of ginseng rhizome could effectively analyze the formation mechanism of “long-necked rhizome”. This project takes PgCLE and PgWOX feedback regulation as a breakthrough to carry out the research on the structure development of ginseng rhizomes. The mechanism of "long-necked rhizome" could be revealed by using both molecular biology and cell biology methods, aiming to provide a theoretical basis for the selection of high quality ginseng.
