申请人前期针对肿瘤下调抗原呈递细胞摄取抗原和表面呈递分子的表达,筛选出肿瘤组织中一类有效抗原,由此开发了一种具备免疫刺激功能的纳米载体疫苗,有效地抑制肿瘤的生长,复发和转移(Advanced Science,2020)。本项目针对前期研究方法在抗原提取和疫苗治疗过程中对正常机体产生不可避免损伤的问题,提出一种对机体无损、非侵入的光调控肿瘤获得性免疫应答疗法。具体研究:(1)验证免疫突触中关键分子MHC class II在光调控肿瘤获得性免疫应答中的作用;(2)探索光调控是如何通过CIITA调节MHC class II的表达,包括搜寻和验证CIITA入核的活化形式、活化位点、被调控的蛋白激酶及其上游信号分子。最终实现挖掘光调控作用到细胞内的直接靶点,解明光调控生物医学效应和分子机制。研究结果将提示具有独特优势的光调控能够调节机体的特异性免疫应答,抑制肿瘤生长,潜在的用于临床肿瘤病人的治疗。

According to the down-regulation of antigen-presenting cells' uptake of antigen and expression of surface presenting molecules by tumor cells, the applicant has previously screened a class of effective antigens from tumor tissue, and developed a nano carrier vaccine with immune stimulation function, which can effectively inhibit the tumor growth, recurrence and metastasis (Advanced Science, 2020). This project aims at the problem that the previous research methods inevitably damage the normal body in the process of antigen extraction and vaccine treatment, and proposes a non-damage and non-invasive light regulation therapy via acquired immune response to treat tumor. Specific studies: (1) To verify the role of MHC class II, a key molecule in immune synapse, in the photoregulation of tumor acquired immune response; (2) To explore how light regulation regulates the expression of MHC class II through CIITA, including searching and verifying the activation form, activation site, regulated protein kinase and its upstream signal molecules of CIITA. Finally, the direct targets in cells are mined for understanding the biomedical effect and molecular mechanism of light regulation. The results will suggest that light regulation with unique advantages can regulate the acquired immune response of the host to inhibit tumor growth, and potentially be used in the treatment of clinical tumor patients.
