SINE是哺乳动物基因组上分布最广泛的转座子,其插入多态分子标记具有多态性高、分布广和开发成本低等特点,在QTL定位和基因组选择等研究中具有较高的应用价值。且研究证实SINE通过多种方式对基因活性发挥调控作用,但目前对SINE影响基因活性的调控机制研究仍然不足。本人前期研究证实SINE在蛋白编码基因mRNA 3'UTR区具有明显插入偏好性,进一步通过比较基因组学研究,在蛋白编码基因3'UTR区鉴定到196个SINE插入多态位点。在此基础上,本研究拟利用196个多态位点,对其与重要经济性状进行关联分析,获得与重要经济性状显著相关的位点,进而筛选若干位点,分析SINE插入多态对靶基因活性和转录本结构等的影响,并从表观遗传、增强子引入、mRNA稳定性等方面探究其对靶基因转录、翻译及表型影响的分子机制。预期将发表研究论文2-3篇,申请专利1项以上,并为猪分子育种提供若干有价值的分子标记。
英文摘要
SINE is the most widely distributed transposon on the mammalian genome. The markers developed based on SINE insertion polymorphic have the features of high polymorphism, wide distribution and low cost, and show greater application value in studies such as genome selection and QTL mapping. It plays an important role in controlling gene expression. However, the current studies on the mechanism of its influence on gene activity are still inadequate. Our previous research found that SINE have a clear preference for insertion in the 3'UTR region of the protein-coding gene, and the further comparative genomic study found 196 SINE polymorphic insertion sites in the 3'UTR. Line with previous studies, this analysis aims to use the 196 SINE polymorphic insertion sites to analyze their connection with important economic traits, and to pick several sites to analyze the impacts on transcript structure and gene activity. The enhancer induction, molecular mechanisms of epigenetic and stability of mRNA have been explored to study the effect on the transcription, translation, and phenotype of the target gene. Two to three research papers are expected to be published, apply for more than one patent and provide several useful molecular markers for pig molecular breeding.
