肉鸡胫骨软骨发育不良(TD)是一种在快速生长家禽中较为常见的软骨代谢障碍性疾病,给家禽生产造成严重的经济损失。我们证实福美双诱导的TD肉鸡软骨细胞中miR-181b表达显著下调,β-catenin磷酸化水平增高,Wnt/β-catenin通路下游调控受到抑制,伴随软骨细胞异常发育。但miR-181b是否通过调控Wnt/β-catenin通路影响软骨细胞发育,目前尚未清楚。本项目拟以原代软骨细胞为模型,采用miR-181b及其靶基因WIF1过表达或干扰、Wnt/β-catenin通路激活或抑制等手段,检测GSK-3β、PKC、β-catenin和软骨细胞发育相关因子ALP、COL2a1、Aggrecan的表达水平、以及软骨细胞的增殖、分化情况,阐明miR-181b调控Wnt/β-catenin通路对TD肉鸡胫骨软骨细胞发育的关键作用及其调控机制,为肉鸡TD防控提供理论基础。

Tibial chondrodysplasia (TD) is one of the common metabolic disease of cartilage in fast growing poultry birds, which cause serious economic loss to broiler industry. Our previous study found that the expression of miR-181b was significantly down-regulated, while p-β-catenin was increased and Wnt/β-catenin signaling pathways was inhibited in tibial chondrocytes of TD chickens with the abnormal development of chondrocytes. Therefore, it is important to know that how miR-181b regulates the expression of Wnt/β-catenin pathway to affect the development of chondrocytes in TD chickens. So, primary chondrocytes will be used as the research model, miR-181b and its target genes (WIF1) overexpressing or silencing, activation/inhibition of Wnt/β-catenin pathway will also be used to verify our hypothesis. The expression levels of GSK-3β, PKC, β-catenin and chondrocyte development related factors (ALP, COL2a1 and Aggrecan), as well as the proliferation and differentiation of chondrocytes will be detected to elucidate the molecular mechanism of miR-181b to affect the development of chondrocytes in TD chickens by targeting WIF1 mediated Wnt/β-catenin signaling pathway. It will provide a theoretical basis for further revealing the prevention and control of TD in chickens.
