目前我国老龄化问题日益严重,糖脂代谢紊乱老年患者急剧增多。糖脂代谢紊乱属痰湿范畴,与脾胃功能失调密切相关。姜为最常用的中药和调味料之一,主要取其调理脾胃、治疗痰湿的功效,其防治糖脂代谢紊乱的疗效已为现代研究所证实,但机制尚不清楚。我们最新研究证明,[6]-姜辣素对血循环脂联素(APN)和骨骼肌脂联素受体(AdipoR1)的双重调节在改善衰老相关糖脂代谢紊乱中扮演重要角色。基于此重要发现,本项目将精细剖析姜/[6]-姜辣素如何作用于骨骼肌“APN/AdipoR1轴”这一线路,着重探讨姜/[6]-姜辣素重塑骨骼肌毛细血管内皮屏障、调节APN穿内皮屏障能力;调节脂肪组织APN内分泌和骨骼肌APN自分泌;以及APN/AdipoR1及其介导的AMPK/PGC-1α信号在姜改善糖脂代谢紊乱中的作用等关键环节,揭示姜改善衰老相关糖脂代谢紊乱的分子机制,为姜调理脾胃、防治衰老相关代谢性疾病提供新依据。
英文摘要
Over the past few decades, there has been a dramatic increase in the number of the ageing individuals with metabolic disorders in China. Age is a major risk factor for the development of metabolic disorders, such as insulin resistance (IR) and obesity. Metabolic disorder in glucose and lipid belongs to “Phlegm-dampness” due to dysfunction of spleen and stomach in TCM. Ginger is one of the most commonly used spices and medicinal plants around the world. Ginger is attributed to spleen and stomach channels, functions as a spleen-stomach-regulating herb, and widely used in the formulas for Phlegm-dampness syndrome in TCM. It has been well demonstrated that ginger improves metabolic disorders in glucose and lipid. However, their molecular mechanisms are still unknown. Recently,our research team has found that [6]-gingerol improves age-associated glucose and lipid metabolic disorder via regulating APN/AdipoR1. Based on this important finding, we in the proposed project will investigate how ginger and its prominent active component [6]-gingerol remodel endothelial barrier, regulate APN endocrine from adipose tissue and autocrine of skeletal muscle, and modulate APN/AdipoR1 meadiating AMPK/PGC-1α signaling, to clarify the relationship between “APN/AdipoR1 axis” and “the improvement of metabolic disorders in glucose and lipid by ginger”. The findings of this project will not only supply with molecular pharmacological basis for developing new potent medicine for age-associated metabolic disorder from ginger, but also provide new insight into the spleen and stomach-regulating property of ginger in the prevention and treatment of Phlegm-dampness syndrome. Thus, this project has novelty and significance.
