北京鸭是全球各种白羽肉鸭育种的主要素材。如何降低腹部脂肪含量,保持或者提高北京鸭皮脂含量是北京鸭育种面临的一个重要挑战。我们前期工作证明北京鸭皮下与腹部前脂肪细胞分化能力有显著区别。阐明皮下与腹部脂肪分化差异的遗传基础对北京鸭育种有重要作用。本研究围绕北京鸭皮下、腹部前脂肪细胞分化能力差异的遗传基础开展研究。利用转录组、蛋白质组、ATAC-Seq等技术对不同诱导分化时期的两个前脂肪细胞原代细胞进行相关肉鸭脂肪分化能力相关的转录因子筛选。利用RNAi、过表达、co-IP等不同技术对综合排序靠前的5个候选转录因子进行单基因功能研究,确定影响两个不同脂肪细胞分化能力差异的关键转录因子,并构建1个关键转录因子的表达调控网络。本研究将有助于开发北京鸭脂肪分布控制的育种新技术,有望为阐明北京鸭体脂分布机理提供相关理论基础数据。

Pekin duck as a standard duck breed in the worldwide, is the world's major white feather duck breeding material. Controlling the rational distribution of fat is one of the important breeding objectives of Pekin duck breeding. How to reduce abdominal fat content and maintain or increase Pekin duck sebum content is a great challenge for Pekin duck breeding. Our preliminary work proved that there is a significant difference between the subcutaneous and abdominal preadipocyte differentiation ability in Pekin duck. The genetic basis for elucidating the differences in subcutaneous and abdominal fat differentiation plays an important role in the breeding of Pekin duck and the biology of fat.. In this study, we would focus on molecular regulation mechanisms of the different differentiation abilities of subcutaneous preadipocytes and abdominal preadipocytes in Pekin ducks. Transcriptome, proteome and single-cell ATAC-seq were used to screen candidate differentially expressed transcription factors between two preadipocytes. We would use the RNAi, over-expression and co-IP to systematically investigate the top 5 highest ranking transcriptional factors. By using this strategy, we can identify the key regulatory genes that affect the different differentiation ability between two cell lines. By measuring the transcriptome changes during over-expression and knock-down the related genes in cell lines, and also measuring the RNA-seq, we would construct the high quality regulatory networks of the studied functional transcription factor.. The results of this study will help to develop a new breeding technology for controlling fat distribution of Pekin duck, which is also expected to provide a genetic basis for elucidating mechanism of the body fat distribution in Pekin duck.
